Abstract
Penyakit kardiovaskular menjadi salah satu penyebab kematian terbesar secara global. Penyakit kardiovaskular umumnya diakibatkan penyumbatan dalam pembuluh darah, salah satunya adalah trombosis. Trombosis terjadi ketika sistem fibrinolitik tidak mampu mendegradasi fibrin sehingga terjadi pembentukan trombus atau gumpalan darah. Agen fibrinolitik secara komersial memiliki efek samping yang tidak diinginkan, seperti tingkat spesifisitas terhadap fibrin rendah, menyebabkan alergi dan juga relatif mahal. Oleh karena itu, diperlukan agen fibrinolitik alami untuk menangani masalah koagulasi darah. Pada penelitian ini, dilakukan pengujian pada ekstrak protease dari buah mengkudu (Morinda citrifolia) yang diketahui memiliki enzim proteolitik bromelain-like protein. Hasil penelitian menunjukkan bahwa protease pada buah mengkudu dapat mendegradasi gumpalan darah dengan persentase degradasi mencapai 85,49%, serta dapat mendegradasi euglobulin, dan mendegradasi fibrin secara efektif pada rantai non-crosslinked fibrin α, β, dan γ. Oleh karena itu, Morinda citrifolia memiliki potensi sebagai agen fibrinolitik. / Cardiovascular disease is one of the main causes of death globally. Cardiovascular disease is generally caused by blockage in the blood vessel, such as thrombosis. Thrombosis occurs when the fibrinolytic system is unable to degrade fibrin and thus promote the formation of thrombus or blood clot. Commercially available fibrinolytic agents have unwanted side effects, such as low fibrin specificity, causing allergies and are also relatively expensive. Therefore, natural fibrinolytic agents are needed to treat blood coagulation problems. In this study, the protease extracted from noni (Morinda citrifolia) which is known to have the proteolytic enzyme called bromelain-like protein was tested. The results showed that proteases from noni can degrade blood clots with a degradation percentage reaching 85.49%, and can degrade euglobulin as well as fibrin effectively in non-crosslinked fibrin chains α, β, and γ. Therefore, Morinda citrifolia has the potential as a fibrinolytic agent.
