Abstract

Penyakit kardiovaskular merupakan penyakit gangguan fungsi jantung dan pembuluh darah. Faktor risiko untuk penyakit kardiovaskular meliputi hipertensi, obesitas, gaya hidup tidak sehat, merokok, dan konsumsi alkohol berlebihan. Selain itu, penyakit kardiovaskular juga dapat diakibatkan oleh pembentukan gumpalan darah dalam pembuluh darah yang disebut sebagai trombosis. Salah satu agen terapeutik untuk mengatasi penyakit kardiovaskular adalah enzim potease. Hal ini karena enzim protease dapat dimanfaatkan dalam mendukung proses fibrinolisis. Jahe merupakan salah satu sumber enzim protease yang disebut zingibain. Zingibain merupakan anggota keluarga protease yang sama seperti papain yang berfungsi untuk melunakkan daging. Pada penelitian ini dilakukan pengujian potensi ekstrak jahe merah sebagai agen fibrinolitik, menguji kemampuan ekstrak jahe merah untuk mendegradasi gumpalan darah, kemampuan fibrinolitik ekstrak jahe merah dalam mendegradasi fibrin dan mendegradasi gumpalan euglobulin menggunakan metode SDS-PAGE. Tahapan penelitian yang dilakukan yaitu: (1) mengekstrak jahe merah, (2) kuantifikasi protein menggunakan metode Hartree Lowry, (3) Uji Degradasi Gumpalan Darah, (4) Uji Degradasi Euglobulin, (5) Uji Degradasi Fibrin. Hasil yang didapatkan adalah enzim protease dari ekstrak jahe merah (Zingiber officinale var. Rubrum) berhasil mendegradasi gumpalan darah dan melalui hasil uji degradasi fibrin juga menunjukkan bahwa protease dari jahe merah dapat memotong rantai non-crosslinked fibrin dan euglobulin, sehingga jahe merah memiliki aktivitas fibrinolitik oleh enzim proteolitik dan berpotensi sebagai agen fibrinolitik./Cardiovascular disease is a disease of impaired heart and blood vessel function. Risk factors for cardiovascular disease include hypertension, obesity, unhealthy lifestyle, smoking and excessive alcohol consumption. In addition, cardiovascular disease can also be caused by the formation of blood clots in blood vessels, which is called thrombosis. One of the therapeutic agents to overcome cardiovascular disease is the protease enzyme. This is because the protease enzyme can be used to support the process of fibrinolysis. Ginger is a source of a protease enzyme called zingibain. Zingibain is a member of the same protease family as papain which functions to tenderize meat. In this study, the potency of red ginger extract as a fibrinolytic agent was tested, the ability of red ginger extract to degrade blood clots, the ability of red ginger extract to degrade fibrin and euglobulin clot using the SDS- PAGE method. The stages of the research were: (1) the extraction of red ginger, (2) protein quantification using the Hartree Lowry method, (3) Blood Clot Degradation Assay, (4) Euglobulin Degradation Assay, (5) Fibrin Degradation Assay. The results obtained were that the protease enzyme from red ginger extract (Zingiber officinale var. Rubrum) succeeded in degrading blood clots and through the fibrin degradation test results also showed that protease from red ginger could cut non-crosslinked fibrin and euglobulin chains, so that red ginger has fibrinolytic activity by proteolytic enzymes and has the potential as a fibrinolytic agent.
