Abstract
Diabetes melitus merupakan penyakit gangguan metabolisme yang ditandai
dengan tingginya kadar gula darah dan penurunan sekresi insulin. Tanaman
merupakan sumber bahan baku obat yang dapat dijadikan sebagai alternatif
pengobatan diabetes. Amerika Selatan, Amerika Tengah, dan Cina secara
tradisonal tanaman ketul (Bidens pilosa L) digunakan mengobati berbagai penyakit,
salah satunya sebagai antidiabetes. Pada penelitian sebelumnya diketahui daun
ketul mengandung senyawa polifenol, flavonoid, tanin, saponin dan alkaloid yang
berkhasiat sebagai antihiperglikemik. Namun sejauh ini belum ada penelitian yang
lebih lanjut dari daun ketul sebagai antihiperglikemik. Sehingga penelitian ini
bertujuan untuk mengetahui efek antihiperglikemik dan dosis yang efektif dari
ekstrak daun ketul yang diekstraksi dengan metode refluks dan pelarut etanol 70%.
Uji antihiperglikemik dengan metode toleransi glukosa menggunakan hewan uji
mencit 20 ekor yang terbagi dalam 5 kelompok, kelompok negatif (aquadest),
kontrol positif (Glibenklamid), kelompok ekstrak daun ketul dibuat dalam 3 dosis
yaitu; dosis 100 mg/kgBB, dosis 300 mg/kgBB dan dosis 500 mg/kgBB. Hasil yang
diperoleh dari pada ekstrak etanol 70% daun ketul memiliki rendemen sebesar
17,5% dan kadar air sebesar 11,84%. Analisis data dilakukan menggunakan SPPS
versi 26 dengan uji One-Way ANOVA diperoleh hasil terdapat perbedaan yang
bermakna (p < 0,05). Hal tersebut berarti ekstrak etanol 70% daun ketul memiliki
efektivitas yang baik dalam menurunkan kadar glukosa darah dengan dosis yang
efektif 100 mg/kgBB./ Diabetes mellitus is a metabolic disorder characterized by high blood sugar levels
and descreased insulin secretion. Plants are a source of medicinal raw materials
that can be used as an alternative treatment for diabetes. South America, Central
America, and China traditionally the ketul plant (Bidens pilosa L) is used to treat
various diseases, one of which is as an antidiabetic. In previous studies it was
known that ketul leaves contain polyphenolic compounds, flavonoids, tannins,
saponins and alkaloids which are efficacious as antihyperglycemics. But so far
there has been no further research on ketul leaves as an antihyperglycemic agent.
So this study aims to determine the antihyperglycemic effect and the effective dose
of ketul leaf extract extracted by reflux method and 70% ethanol solvent.
Antihyperglicemic test using glucose tolerance method using 20 mice as test animal
divided into 5 groups, negative group (aquadest), positive control (Glibenclamide),
ketul leaf extract group made in 3 doses namely; a dose of 100 mg/kgBB, a dose of
300 mg/kgBB and a dose of 500 mg/kgBB. The results obtained from the 70%
ethanol extract of ketul leaves had a yield of 17,5% and a water content of 11,84%.
Data analysis was performed using SPPS version 26 with the One-Way ANOVA
test. The results showed that there were significant differences (p < 0,05). This
means that the 70% ethanol extract of ketul leaves has good effectiveness in
lowering blood glucose levels with an effective dose of 100 mg/kgBB.
