Abstract
Diabetes merupakan gangguan metabolik kronis yang dengan karakteristik berupa kenaikan kadar glukosa darah di atas normal akibat dari gangguan sekresi dan sensitivitas hormon insulin yang meregulasi gula dalam darah. Salah satu cara pencegahan penyakit diabetes adalah menghambat enzim α-amilase. Enzim α-amilase dapat disekresikan oleh berbagai organisme, termasuk bakteri. Indonesia memiliki sumber daya alam yang melimpah yang dapat digunakan sebagai tumbuhan obat. Penelitian ini bertujuan untuk mengeksplorasi potensi penghambatan enzim α-amilase dari bakteri Bacillus amyloliquefaciens dan Lactobacillus plantarum oleh fraksi Castanopsis tungurrut. Penelitian ini menggunakan metode in vitro dan in silico. Hasil in vitro menunjukkan bahwa fraksi batang Castanopsis tungurrut menghasilkan aktivitas penghambatan dengan nilai IC50 sebesar 11,0514 ppm pada α-amilase dari B. amyloliquefaciens dan 47,0307 ppm untuk α-amilase dari L. plantarum. Untuk hasil in silico dengan menggunakan molecular docking menunjukkan bahwa senyawa picrasinoside A dan mururin A memiliki potensial untuk dikembangkan menjadi suatu obat./Diabetes is a chronic metabolic disorder characterized by elevated blood glucose levels above normal due to impaired secretion and sensitivity of the insulin hormone, which regulates blood sugar. One way to prevent diabetes is by inhibiting the α-amylase enzyme. α-amylase enzyme is secreted by various organisms, one of which is bacteria. Indonesia has abundant natural resources that can be used as medicinal plants. This study aims to explore the potential inhibition of α-amylase enzyme from bacteria Bacillus sp. and Lactobacillus sp. by Castanopsis tungurrut fraction. This study was carried out by in vitro and in silico method. In vitro results showed that the Castanopsis tungurrut stem fraction produced inhibitory activity with an IC50 value of 11.0514 ppm for α-amylase from B. amyloliquefaciens and 47.0307 ppm for α-amylase from L. plantarum. In silico results using molecular docking show that the compounds picrasinoside A and mururin A have the potential to be developed into a drug.
