Abstract
Jerawat merupakan salah satu penyakit kulit yang umum terjadi pada remaja. Bakteri penyebab jerawat antara lain Propionibacterium acnes dan Staphylococcus aureus. Pengobatan jerawat dapat dilakukan dengan pemberian antibiotik, namun penggunaan yang tidak tepat dapat menyebabkan resistensi. Salah satu upaya untuk meminimalisir resistensi antibiotik yaitu dengan penggunaan bahan alam. Daun pegagan mengandung saponin dan daun mimba mengandung flavonoid yang memiliki aktivitas antibakteri terhadap bakteri penyebab jerawat. Penelitian ini bertujuan untuk mengetahui Konsentrasi Hambat Minimum (KHM) dan Konsentrasi Bunuh Minimum (KBM) terhadap bakteri Propionibacterium acnes dan Staphylococcus aureus. Daun pegagan dan daun mimba dimaserasi dengan pelarut etanol 70%. Pengujian aktivitas antibakteri menggunakan metode mikrodilusi cair untuk nilai KHM dan metode dilusi padat untuk nilai KBM. Kombinasi ekstrak daun pegagan dan daun mimba diharapkan dapat meningkatkan efektivitas dalam menghambat dan membunuh bakteri Propionibacterium acnes dan Staphylococcus aureus. Dari hasil penelitian didapatkan nilai KHM90 ekstrak kombinasi dengan konsentrasi 10000 ppm + 312,5 ppm terhadap bakteri Propionibacterium acnes dan konsentrasi 312,5 ppm + 10000 ppm terhadap bakteri Staphylococcus aureus. / Acne is one of the most common skin problems among teenagers. Bacteria that cause acne include Propionibacterium acnes and Staphylococcus aureus. Antibiotics can be used to treat acne, however incorrect use may cause resistance. The usage of natural materials is one of the measures to reduce antibiotic resistance. Saponins that found in pegagan leaves, and flavonoids in mimba leaves show antibacterial effect against acne-causing bacteria. This study aims to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) against Propionibacterium acnes and Staphylococcus aureus bacteria. Pegagan and mimba leaves were macerated with 70% ethanol solvent. Antibacterial activity testing used liquid microdilution method for KHM value and solid dilution method for KBM value. The combination of pegagan and mimba leaf extracts is expected to increase the effectiveness of inhibiting and killing Propionibacterium acnes and Staphylococcus aureus bacteria. From the results of the study the MIC90 values combination extract with a concentration 10000 ppm + 312,5 ppm against Propionibacterium acnes bacteria and concentration 312,5 ppm + 10000 ppm against Staphylococcus aureus bacteria.
