Abstract
Bifidobacterium merupakan bakteri yang umum ditemukan dalam usus manusia dan berperan dalam menjaga serta memberikan berbagai manfaat kesehatan seperti mencegah infeksi bakteri dan virus, mencegah inflamasi, mencegah kanker, dan mendukung kesehatan psikologis. Berbagai penelitian telah menunjukkan bahwa rafinosa berupa trisakarida yang ditemukan dalam kacang polong, kacang kedelai, serta biji, akar, dan daun tanaman legum dapat mendukung pertumbuhan Bifidobacterium dan menunjukkan potensinya sebagai prebiotik. Penelitian ini bertujuan untuk menganalisis kemampuan fermentasi rafinosa serta kemampuan pertumbuhan Bifidobacterium breve BS2-PS2 dan Bifidobacterium animalis BR2-5 setelah suplementasi rafinosa melalui uji aktivitas fermentasi dan uji pertumbuhan. Uji aktivitas fermentasi dilakukan dengan menumbuhkan Bifidobacterium dalam TPY broth yang disuplementasi rafinosa 2% dalam keadaan anaerobik selama 72 jam. Kemudian uji pertumbuhan dilakukan dengan menginokulasi sampel uji aktivitas fermentasi pada jam ke-0, 24, 48, dan 72 pada TPY agar dan diinkubasi selama 72 jam dalam keadaan anaerobik. Hasil uji aktivitas fermentasi menunjukkan bahwa kedua strain Bifidobacterium yang digunakan dapat memfermentasi rafinosa yang ditunjukkan melalui penurunan pH medium yang tinggi, dari pH awal 7.4 ke pH 4.91 (SD = 0.12) untuk B. breve BS2-PS2 dan pH 4.90 untuk B. animalis BR2-5. Hasil uji pertumbuhan menunjukkan tanda-tanda peningkatan pertumbuhan setelah suplementasi rafinosa, tetapi hasil belum dapat ditentukan dengan pasti. Penelitian lebih lanjut masih perlu dilakukan untuk menentukan kemampuan rafinosa dalam meningkatkan pertumbuhan B. breve BS2-PS2 dan B. animalis BR2-5. / Bifidobacterium is a bacteria commonly found in the human intestine and plays a role in maintaining and providing various health benefits such as preventing bacterial and viral infections, preventing inflammation, preventing cancer, and supporting psychological health. Various studies have shown that raffinose, a trisaccharide found in peas, soybeans, and the seeds, roots and leaves of legume plants, can support the growth of Bifidobacterium and show its potential as a prebiotic. This study aims to analyze the fermentation ability of raffinose and the growth ability of Bifidobacterium breve BS2-PS2 and Bifidobacterium animalis BR2-5 after raffinose supplementation through fermentation activity assay and growth test. The fermentation activity assay was carried out by growing Bifidobacterium in TPY broth supplemented with 2% raffinose under anaerobic conditions for 72 hours. Then the growth test was carried out by inoculating the fermentation activity assay samples at 0, 24, 48 and 72 hours on TPY agar and incubating for 72 hours under anaerobic conditions. The results of the fermentation activity assay showed that the two Bifidobacterium strains used were able to ferment raffinose as indicated by a high decrease in the pH of the medium, from initial pH 7.4 to pH 4.91 (SD = 0.12) for B. breve BS2-PS2 and pH 4.90 for B. animalis BR2 -5. Growth test results show signs of increased growth after raffinose supplementation, but the results cannot be determined with certainty. Further research still needs to be carried out to determine the ability of raffinose to increase the growth of B. breve BS2-PS2 and B. animalis BR2-5.
