Abstract

Salah satu spesies yang telah banyak dipelajari karakteristiknya sebagai bakteri probiotik dengan berbagai manfaat kesehatan bagi tubuh merupakan Bifidobacterium animalis. Salah satu kriteria seleksi bagi bakteri probiotik merupakan kemampuannya untuk melekat pada jaringan sel epitel. Kemampuan bakteri probiotik untuk melekat pada dinding usus tersebut dinamakan sebagai adhesi bakteri. Musin memiliki peran penting dalam kolonisasi gastrointestinal mikroba karena bakteri probiotik maupun patogen harus dapat menempel pada musin. Musin yang digunakan dalam penelitian merupakan musin komersial tipe II. Penelitian ini berfokus pada uji adhesi Bifidobacterium animalis strain BR2-5 yang dengan tujuan untuk mengetahui seberapa besar kemampuan BR2-5 menempel pada musin dan kemudian dibandingkan dengan hasil persentase adhesi BR2-5 pada microplates tanpa musin sebagai kontrol negatif dan Lactiplantibacillus plantarum SUKC-1a sebagai kontrol positif. Akan dilakukan juga pengukuran konsentrasi protein larutan musin dengan metode persiapan yang berbeda dengan tujuan mendapatkan larutan musin dengan konsentrasi akhir yang tinggi. Persiapan larutan musin dilakukan dengan 5 metode yang berbeda dan hasil pengukuran Bradford assay menunjukkan metode ke-empat (M4) dan ke-lima (M5) memiliki konsentrasi protein yang cukup untuk digunakan dalam uji adhesi yaitu 0,596 mg/mL pada M4 dan 1,099 mg/mL pada M5. Hasil pengujian adhesi menunjukkan adanya kemampuan B. animalis strain BR2-5 untuk menempel pada porcine mucin type-II dengan nilai adhesi tertinggi 7,37 ± 2,40 jika dibandingkan dengan hasil kontrol negatif sebesar 0,33 ± 0,51. Uji adhesi kontrol positif menggunakan Lb. plantarum SUKC-1a memiliki persentase adhesi yang setara dengan BR2-5 dengan rata-rata hasil tertinggi sebesar 9,64%. Dari hasil uji adhesi yang telah dilakukan, dapat disimpulkan B. animalis BR2-5 memiliki salah satu karakteristik sebagai bakteri probiotik berpotensial. / One of the species that has been extensively studied for its characteristics as a probiotic microorganism with various health benefits for the body is Bifidobacterium animalis. One of the selection criteria for probiotic bacteria is their ability to adhere to epithelial cell tissues. This ability of probiotic bacteria to adhere to the intestinal wall is known as bacterial adhesion. Mucin plays an important role in the colonization of gastrointestinal microbes because both probiotic and pathogenic bacteria must be able to adhere to mucin. The mucin used in this research is commercially available type II mucin. This study focuses on the adhesion assay of Bifidobacterium animalis strain BR2-5 with the aim of determining the adhesion capability of BR2-5 to mucin and then comparing the adhesion percentage of BR2-5 to microplates without mucin as a negative control and Lactiplantibacillus plantarum SUKC-1a as a positive control. The protein concentration of mucin swill also be measured using different preparation methods to obtain a mucin solution with a high final concentration. The mucin solution was prepared 5 times using different methods, and the Bradford assay results showed that the fourth (M4) and fifth (M5) methods had sufficient protein concentrations for the adhesion test, with 0.596 mg/mL for M4 and 1.099 mg/mL for M5. The adhesion test results showed that the BR2-5 strain was able to adhere to porcine mucin type-II with the highest adhesion value of 7.37 ± 2.40 compared to the negative control result of 0.33 ± 0.51. The positive control adhesion test using Lb. plantarum SUKC-1a had an equivalent adhesion percentage to BR2-5 with the highest average result of 9,64%. From the adhesion test results, it can be concluded that B. animalis BR2-5 has one of the characteristics of a potential probiotic bacteria.
