Abstract
Radikal bebas dapat dibentuk melalui radiasi sinar ultraviolet, minuman keras, rokok, polusi udara, pestisida dan olahan makanan yang tidak berkhasiat. Radikal
bebas juga dapat menyebabkan berbagai masalah kesehatan seperti penuaan dini, penyakit asma, penyakit diabetes melitus serta penyakit kanker. Senyawa yang dapat mencegah kerusakan sel dan dapat mempertahankan tubuh dari radikal bebas yaitu berupa antioksidan. Salah satu antioksidan yang dapat diperoleh dalam bentuk alami dan memiliki sumber potensial adalah tanaman melinjo (Gnetum gnemon L.). Penelitian ini bertujuan untuk mengetahui senyawa metabolit sekunder dan aktivitas antioksidan ekstrak etanol 96% dan diklorometana kulit melinjo merah (Gnetum gnemon L.) berdasarkan parameter IC50. Penelitian ini meliputi tahap pembuatan simplisia, pembuatan ekstrak dengan pelarut etanol 96% dan diklorometana dengan metode maserasi, pengujian skrining fitokimia, pengukuran aktivitas antioksidan dengan metode DPPH menggunakan spektrofotometer UV-Vis. Analisis data dilakukan dengan menggunakan nilai % inhibisi dan persamaan linier dengan membuat kurva kalibrasi dan nilai IC50. Data dianlisis dengan membuat kurva kalibrasi sehingga mendapatkan persamaan regresi linear dan menghitung nilai IC50. Hasil uji skrining fitokimia yang telah diperoleh pada ekstrak etanol 96% dan ekstrak diklorometana kulit melinjo (Gnetum gnemon L.) yang diketahui mengandung senyawa metabolit sekunder meliputi alkaloid, flavonoid, saponin, dan steroid/terpenoid. Hasil pengujian aktivitas antioksidan ekstrak etanol 96% dan ekstrak diklorometana diketahui bahwa kulit melinjo (Gnetum gnemon L.) memiliki aktivitas antioksidan yang tidak aktif dengan nilai IC50 masing-masing sebesar 455,2355 ppm dan 1144,6806 ppm/Free radicals can be formed through ultraviolet radiation, alcohol, cigarettes, air pollution, pesticides and processed foods that are not nutritious. Free radicals can also cause various health problems such as premature aging, asthma, diabetes mellitus and cancer. Compounds that can prevent cell damage and can defend the body from free radicals are called antioxidant. One of the source of antioxidant that can be obtained in natural form and has a high potential is the melinjo plant (Gnetum gnemon L.). This study aims to determine the secondary metabolites compounds and to conduct antioxidant activity test of 96% ethanol and dichloromethane extracts of red melinjo peel (Gnetum gnemon L.) based on IC50 parameters. This research includes the preparation of simplicia, making extracts with 96% ethanol and dichloromethane solvents by maceration method, phytochemical screening testing, measuring antioxidant activity with DPPH method using UV-Vis spectrophotometer. Data analysis was done using % inhibition value and linear equation by making calibration curve and IC50 value. The data is analyzed by creating a calibration curve so as to obtain a linear regression equation and calculate the IC50 value. The results of phytochemical screening tests that have been obtained on 96% ethanol extract and melinjo bark dichloromethane extract (Gnetum gnemon L.) which are known to contain secondary metabolite compounds include alkaloids, flavonoids, saponins, and steroids / terpenoids. The results of testing the antioxidant activity of 96% ethanol extract and dichloromethane extract found that melinjo peel (Gnetum gnemon L.) has inactive antioxidant activity with IC50 values of 455.2355 ppm and 1144.6806 ppm respectively.
