Abstract
Penelitian ini melanjutkan penelitian oleh Napitupulu (2023) tentang ekstrak etil
asetat daun pepaya jepang (Cnidoscolus aconitifolius (Mill.) I.M.Johnst.) terhadap
S. aureus, dengan memodifikasi bakteri menjadi E. coli penyebab diare dan uji
KHM. Karena tidak ada daya hambat terhadap E. coli, bakteri dimodifikasi menjadi
S. epidermidis. Namun, masalah teknis menyebabkan pengujian dilanjutkan dengan
menggunakan ekstrak etanol 96% daun pepaya jepang terhadap S. epidermidis
penyebab jerawat. Hasil skrining ekstrak etanol 96% daun pepaya jepang
mengandung senyawa alkaloid, flavonoid, fenol, saponin, dan tanin sebagai agen
antibakteri. Penelitian ini bertujuan mengetahui zona hambat yang terbentuk dari
aktivitas antibakteri ekstrak etanol 96% daun pepaya jepang terhadap
Staphylococcus epidermidis penyebab jerawat dan uji KHM. Metode yang
digunakan adalah difusi cakram dengan tetrasiklin sebagai kontrol positif.
Konsentrasi ekstrak yang mampu menghambat bakteri yaitu 1.000.000 ppm,
750.000 ppm, dan 500.000 ppm dengan daya hambat rata-rata berturut-turut 12,6
mm; 9,78 mm; dan 8,42 mm (semuanya resistent). Uji KHM menunjukkan daya
hambat terendah pada konsentrasi 500.000 ppm dengan persentase inhibisi
143,552%. Analisis data menunjukkan perbedaan signifikan dengan uji pos hoc
Tamhane’s / This study continues research by Napitupulu (2023) on ethyl acetate extract of
Japanese papaya leaves (Cnidoscolus aconitifolius (Mill.) I.M.Johnst.) against S.
aureus, by modifying the bacteria into E. coli causing diarrhea and KHM test. Since
there was no inhibition against E. coli, the bacteria were modified into S.
epidermidis. However, technical problems caused the test to continue using 96%
ethanol extract of Japanese papaya leaves against S. epidermidis that causes acne.
Screening results of 96% ethanol extract of Japanese papaya leaves contain
alkaloid, flavonoid, phenol, saponin, and tannin compounds as antibacterial
agents. This study aims to determine the inhibition zone formed from the
antibacterial activity of 96% ethanol extract of Japanese papaya leaves against
Staphylococcus epidermidis causing acne and KHM test. The method used was disc
diffusion with tetracycline as a positive control. Concentrations of extracts that are
able to inhibit bacteria are 1,000,000 ppm, 750,000 ppm, and 500,000 ppm with an
average inhibition of 12.6 mm; 9.78 mm; and 8.42 mm, respectively (all are
resistant). The KHM test showed the lowest inhibition at a concentration of 500,000
ppm with a percentage inhibition of 143.552%. Data analysis showed significant
differences with Tamhane's post hoc test.
