Abstract
Jerawat merupakan masalah kulit yang terjadi karena peradangan pada folikel polisebasea. Salah satu bakteri penyebab infeksi jerawat adalah Staphylococcus epidermidis, yang terdapat pada kulit dan selaput lendir manusia. Mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl.) merupakan tumbuhan asli Papua, Indonesia. Daun mahkota dewa mengandung senyawa metabolit sekunder alkaoid, saponin, dan tanin yang berfungsi sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari ekstrak etanol 96% daun mahkota dewa terhadap bakteri Staphylococcus epidermidis dengan metode difusi cakram serta menentukan Konsentrasi Hambat Minimum (KHM) menggunakan spektrofotometer UV-Vis. Rendemen ekstrak etanol 96% daun mahkota dewa diperoleh sebesar 23,285%, dan kadar air ekstrak 7,81%. Hasil skrining fitokimia daun mahkota dewa mengandung senyawa metabolit sekunder alkaloid, flavonoid, tanin, saponin, fenol, steroid, dan kuinon. Penelitian menunjukkan daya hambat sebesar 8,98 mm; 10,24 mm; dan 13,14 mm pada konsentrasi ekstrak 100.000 ppm, 250.000 ppm, dan 500.000 ppm. Konsentrasi 100.000 ppm, dan 250.000 ppm menunjukkan zona hambat sedang, sedangkan konsentrasi 500.000 ppm menunjukkan zona hambat kuat terhadap pertumbuhan bakteri Staphylococcus epidermidis. Daya hambat yang dihasilkan tidak lebih besar dari tetrasiklin. Semakin besar konsentrasi yang digunakan, semakin besar zona hambat yang terbentuk. KHM ekstrak etanol 96% daun mahkota dewa adalah pada konsentrasi 6.250 ppm dengan % IR sebesar 8,314%. / Acne is a skin problem that occurs due to inflammation of the polysebaceous follicles. One of the bacterial causes of acne infection is Staphylococcus epidermidis, which is found on human skin and mucous membranes. Mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl.) is a plant native to Papua, Indonesia. Mahkota dewa leaves contain secondary metabolite compounds of alkaoids, saponins and tannins which function as antibacterials. This research aims to determine the antibacterial activity of 96% ethanol extract of Mahkota dewa leaves against Staphylococcus epidermidis bacteria using the disc diffusion method and determine the Minimum Inhibitory Concentration (MIC) using a UV-Vis spectrophotometer. The yield of 96% ethanol extract of Mahkota dewa leaves was 23.285%, and the water content of the extract was 7.81%. The results of the phytochemical screening of Mahkota dewa leaves contain secondary metabolite compounds of alkaloids, flavonoids, tannins, saponins, phenols, steroids and quinones. Research shows a resistance of 8.98 mm; 10.24mm; and 13.14 mm at extract concentrations of 100,000 ppm, 250,000 ppm, and 500,000 ppm. Concentrations of 100,000 ppm and 250,000 ppm indicate a moderate inhibition zone, while a concentration of 500,000 ppm indicates a strong inhibition zone against the growth of Staphylococcus epidermidis bacteria. The resulting inhibitory power is not greater than tetracycline. The greater the concentration used, the greater the inhibition zone formed. The MIC of 96% ethanol extract of Mahkota dewa leaves is at a concentration of 6,250 ppm with an IR% of 8.314%.
