Abstract

Tanaman biwa (Eriobotrya japonica (Thunb.) Lindl) diketahui mengandung senyawa flavonoid, tanin, glikosida, saponin dan terpenoid yang berperan dalam aktivitas antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri fraksi aktif kulit batang dan mengetahui sediaan krim fraksi yang memenuhi persyaratan krim yang baik. Penelitian ini meliputi pembuatan simplisia dan ekstrak, fraksinasi ekstrak, skrining fitokimia fraksi, uji antibakteri fraksi terhadap bakteri Staphylococcus aureus, pembuatan krim fraksi aktif kulit batang biwa dan evaluasi krim yang meliputi organoleptik, homogenitas, uji pH, uji daya sebar, uji daya lekat dan viskositas. Pengujian antibakteri menggunakan metode cakram dengan beberapa sampel yaitu fraksi ekstrak kulit batang biwa konsentrasi 500.000 ppm, 100.000 ppm, dan 10.000 ppm, kontrol positif eritromisin 15µg dan kontrol negatif DMSO 10%. Dua dari ketiga fraksi yang di uji memiliki aktivitas antibakteri yaitu fraksi etanol dan etil setat sedangkan fraksi n-heksan tidak memiliki aktivitas antibakteri. Uji aktivitas antibakteri fraksi etanol menghasilkan zona hambat 23,31 mm (500.000ppm) kategori sangat kuat, 16,81mm (100.000ppm) kategori kuat, dan 14,18mm (10.000ppm) kategori kuat sedangkan fraksi etil asetat menghasilkan zona hambat 20,93 mm (500.000ppm) kategori sangat kuat, 15,12 mm (100.000ppm) kategori kuat, dan 13,37 mm (10.000ppm) kategori kuat. Sediaan krim dibuat dengan fraksi etanol dan etil asetat dengan konsentrasi 1% yang kemudian dilakukan evaluasi serta uji stabilitas/Loquat plants (Eriobotrya japonica (Thunb.) Lindl) are known to contain flavonoids, tannins, glycosides, saponins, and terpenoids,which play a role in antibacterial activity. This study aims to determine the antibacterial activity of the active fraction of stem bark and determine the cream fraction preparation that meets the requirements for a good cream. This research includes making simplicia and extracts, fractionating extracts, phytochemical screening of fractions, antibacterial testing of fractions against Staphylococcus aureus bacteria, making active cream fractions of loquat bark, and evaluating creams, which include organoleptic, homogeneity, pH tests, spreadability tests, adhesion tests, and viscosity. Antibacterial testing used the disc method with several samples, namely the loquat bark extract fraction with concentrations of 500,000 ppm, 100,000 ppm, and 10,000 ppm, a positive control of 15 µg erythromycin, and a negative control of 10% DMSO. Two of the three fractions tested had antibacterial activity, namely the ethanol and ethyl cetate fractions, while the n-hexane fraction did not have antibacterial activity. The antibacterial activity test of the ethanol fraction produced an inhibition zone of 23.31 mm (500,000 ppm) in the very strong category, 16.81 mm (100,000 ppm) in the strong category, and 14.18 mm (10,000 ppm) in the strong category, while the ethyl acetate fraction produced an inhibition zone of 20.93 mm (500,000 ppm) in the in the very strong category, 15.12 mm (100,000 ppm) in the in the strong category, and 13.37 mm (10,000 ppm) in the in the strong category. The cream preparation was made from ethanol and ethyl acetate fractions with a concentration of 1%, which was then evaluated and tested for stability.
