Abstract
Masyarakat Indonesia seringkali terpapar polusi udara dari lingkungan sekitar saat menjalani kehidupan sehari-hari. Polusi udara berbahaya bagi kesehatan yang dapat menyebabkan radikal bebas dalam tubuh. Efek radikal bebas membentuk stres oksidatif dan menjadi penyebab terjadinya penyakit lainnya. Senyawa yang bersifat antioksidan mampu menghambat stres oksidatif dan mampu menghambat pembentukan radikal bebas. Salah satu tanaman herbal yang sering dikenal memiliki efek antioksidan adalah rosemary. Penelitian ini dilakukan dengan tujuan melihat aktivitas antioksidan ekstrak etanol 96% dan etil asetat daun rosemary. Serta uji total kandungan flavonoid-fenolik pada ekstrak etanol 96% dan etil asetat daun rosemary. Metode penelitian ini menggunakan DPPH untuk menentukan aktivitas antioksidan ekstrak. Uji kandungan flavonoid akan ditentukan dengan reagen AlCl3 sedangkan uji kandungan fenolik menggunakan reagen Folin-Ciocalteau. Hasil uji antioksidan ekstrak etanol 96% memiliki IC50 48,519 ppm dan ekstrak etil asetat dengan IC50 47,493 ppm. Hasil uji kandungan flavonoid ekstrak etanol 96% adalah 68,724 mgQE/g dan ekstrak etil asetat adalah 44,008 mgQE/g. Hasil uji kandungan fenolik ekstrak etanol 96% adalah 138,773 mgGAE/g dan ekstrak etil asetat adalah 166,928 mgGAE/g. Kesimpulan yang didapat adalah daun rosemary memiliki aktivitas antioksidan yang kuat dan ditemukan total kandungan flavonoid dan fenolik pada kedua ekstrak./Indonesian are often exposed to air pollution and it’s dangerous because it can cause free radicals inside the body. The effects of free radicals form oxidative stress and cause other diseases. Antioxidant compounds are able to inhibit oxidative stress and the formation of free radicals. The rosemary plant is known to have antioxidant effects. This research was conducted with the aim of examining the antioxidant activity of 96% ethanol extract and ethyl acetate of rosemary leaves. Also test the total flavonoid-phenolic content in both extracts of rosemary leaves. This research method uses DPPH to determine the antioxidant activity. The flavonoid content test will be determined by AlCl3 reagent, while the phenolic content test will use the Folin-Ciocalteau reagent. The antioxidant test results of the 96% ethanol and ethyl acetate extract had an IC50 of 48,519 ppm and 47,493 ppm. The test results for the flavonoid content of 96% ethanol and ethyl acetate extract were 68.724 mgQE/g and 44.008 mgQE/g. The test results for the phenolic content of 96% ethanol and ethyl acetate extract were 138.773 mgGAE/g and 166.928 mgGAE/g. The conclusion obtained that rosemary leaves have strong antioxidant activity and the flavonoid-phenolic content was found in both extracts.
