Abstract
Staphylococcus epidermidis adalah bakteri Gram-positif yang sering menjadi penyebab infeksi nosokomial, terutama pada pasien dengan perangkat medis yang terimplan. Resistensi bakteri ini terhadap antibiotik konvensional mendorong pencarian alternatif baru yang efektif, salah satunya adalah ekstrak tumbuhan. Daun bangun-bangun (Plectranthus amboinicus) diketahui memiliki potensi sebagai agen antibakteri karena kandungan senyawa bioaktifnya. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak etanol 70% daun Bangun-bangun terhadap bakteri Staphylococcus epidermidis. Penelitian ini menggunakan metode maserasi dalam proses ekstraksi, difusi cakram dalam pengujian aktivitas antibakteri dan metode Spektrofotometri UV-Vis dalam pengujian Konsentrasi Hambat Minimum (KHM) bakteri. Hasil penelitian menunjukkan bahwa ekstrak etanol 70% daun bangun-bangun mengandung metabolit sekunder flavonoid, tanin, fenol, kuinon, saponin, triterpenoid, dan steroid. Hasil diameter zona hambat pada konsentrasi uji 500.000 ppm, 50.000 ppm, dan 10.000 ppm masing-masing secara berurutan sebesar 18,11 mm; 14,78 mm; dan 13,93 mm. Konsentrasi Hambat Minimum (KHM) ekstrak etanol 70% daun bangun-bangun adalah sebesar 5.000 ppm. Berdasarkan analisis data dapat disimpulkan bahwa ekstrak etanol 70% daun bangun-bangun memiliki aktivitas antibakteri terhadap Staphylococcus epidermidis dengan konsentrasi uji yang optimal adalah 10.000 ppm./Staphylococcus epidermidis is a Gram-positive bacterium that often causes nosocomial infections, especially in patients with implanted medical devices. The resistance of this bacteria to conventional antibiotics has encouraged the search for new effective alternatives, one of which is plant extracts. Bangun-bangun leaves (Plectranthus amboinicus) are known to have potential as an antibacterial agent because of their bioactive compound content. This study aims to determine the antibacterial activity of 70% ethanol extract of Bangun-bangun leaves against Staphylococcus epidermidis bacteria. This research uses the maceration method in the extraction process, disc diffusion in testing antibacterial activity and the UV-Vis Spectrophotometry method in testing the Minimum Inhibitory Concentration (MIC) of bacteria. The results showed that the 70% ethanol extract of bangun
bangun leaves contained secondary metabolites of flavonoids, tannins, phenols, quinones, saponins, triterpenoids and steroids. The results of the diameter of the inhibition zone at test concentrations of 500,000 ppm, 50,000 ppm, and 10,000 ppm were respectively 18,11 mm;, 14,78 mm; and 13,93 mm. The Minimum Inhibitory Concentration (MIC) of 70% ethanol extract of bangun-bangun leaves is 5,000 ppm. Based on data analysis, it can be concluded that the 70% ethanol extract of bangun-bangun leaves has antibacterial activity against Staphylococcus epidermidis with test concentration being 10,000 ppm.
