Abstract

Jerawat adalah kondisi kulit yang terjadi karena penumpukan minyak di kulit wajah, yang menyebabkan pori-pori tersumbat dan memicu peradangan serta pertumbuhan bakteri. Jenis bakteri seperti Propionibacterium acnes, Staphylococcus aureus, dan Staphylococcus epidermidis merupakan jenis bakteri yang umumnya dapat menyebabkan jerawat. Daun jeruk purut mempunyai senyawa metabolit sekunder yaitu flavonoid, tanin, dan saponin. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antibakteri ekstrak etil asetat daun jeruk purut dan menentukan nilai KHM serta KBM dari ekstrak etil asetat daun jeruk purut terhadap bakteri Propionibacterium acnes dan Staphylococcus epidermidis. Ekstrak etil asetat daun jeruk purut akan didapatkan dengan metode maserasi dan dibuat dalam 3 seri konsentrasi. Penentuan aktivitas antibakteri menggunakan metode sumuran dengan tetrasiklin dan vankomisin sebagai kontrol positif dan DMSO sebagai kontrol negatif. Penentuan nilai KBM menggunakan metode spread plate dan untuk menentukan nilai KHM akan diukur menggunakan spektrofotometri UV-Vis. Zona hambat terhadap bakteri Propionibacterium acnes dan Staphylococcus epidermidis pada konsentrasi 50.000, 25.000, dan 12.500 secara berturut-turut yaitu 17 mm; 16,8 mm; 15,5 mm dan 22,7 mm; 16,2 mm; 15,8 mm. Konsentrasi Hambat Minimum bakteri Propionibacterium acnes pada konsentrasi 50.000 ppm dan bakteri Staphylococcus epidermidis pada konsentrasi 100.000 ppm dan 50.000. Ekstrak tidak didapatkan hasil nilai Konsentrasi Bunuh Minimum karena pertumbuhan bakteri masih terlihat. Dapat disimpulkan ekstrak etil asetat daun jeruk purut mempunyai aktivitas antibakteri pada bakteri Staphylococcus epidermidis dan pada bakteri Propionibacterium acnes hanya konsentrasi terkecil yang tidak mempunyai aktivitas antibakteri. Bakteri Staphylococcus epidermidis mempunyai nilai KHM pada setiap konsentrasi. Bakteri Propionibacterium acnes mempunyai nilai KHM pada konsentrasi 25.000 ppm, 6.250 ppm, dan 3.125. Nilai KBM pada kedua bakteri tidak didapatkan./Acne is a skin condition that occurs due to a build-up of oil on the facial skin, which clogs the pores and triggers inflammation and bacterial growth. Types of bacteria such as Propionibacterium acnes, Staphylococcus aureus, and Staphylococcus epidermidis are types of bacteria that can commonly cause acne. Kaffir lime leaves have secondary metabolite compounds, namely flavonoids, tannins, and saponins. The purpose of this study was to determine the antibacterial activity of ethyl acetate extract of kaffir lime leaves and determine the KHM and KBM values of ethyl acetate extract of kaffir lime leaves against Propionibacterium acnes and Staphylococcus epidermidis bacteria. The ethyl acetate extract of kaffir lime leaves will be obtained by maceration method and made in 3 concentration series. Determination of antibacterial activity using the pitting method with tetracycline and vancomycin as positive controls and DMSO as negative control. Determination of KBM value using spread plate method and to determine KHM value will be measured using UV-Vis spectrophotometry. The zone of inhibition against Propionibacterium acnes and Staphylococcus epidermidis bacteria at concentrations of 50,000, 25,000, and 12,500 were 17 mm; 16.8 mm; 15.5 mm and 22.7 mm; 16.2 mm; 15.8 mm, respectively. Minimum inhibitory concentration of Propionibacterium acnes bacteria at a concentration of 50,000 ppm and Staphylococcus epidermidis bacteria at a concentration of 100.000 ppm and 50.000 ppm. The extract did not have a Minimum Kill Concentration value because bacterial growth was still visible. It can be concluded that ethyl acetate extract of kaffir lime leaves has antibacterial activity on Staphylococcus epidermidis bacteria and on Propionibacterium acnes bacteria only the smallest concentration does not have antibacterial activity. Staphylococcus epidermidis bacteria have KHM values at each concentration. Propionibacterium acnes bacteria have KHM values at concentrations of 25,000 ppm, 6,250 ppm, and 3,125. KBM values for both bacteria were not obtained.
