Abstract

Radikal bebas adalah zat asing yang dapat menyusup ke dalam tubuh manusia dan berpotensi merusak sistem imun. Antioksidan adalah senyawa yang mampu menghambat atau menghalangi terbentuknya radikal bebas. Secara alami, tubuh telah memiliki antioksidan, tetapi jumlahnya sering kali tidak mencukupi dalam mencegah radikal bebas berlebih. Maka dari itu, diperlukan antioksidan eksogen. Salah satu tumbuhan yang memiliki antioksidan alami adalah genus Ficus. Penelitian ini bertujuan untuk menentukan fraksi, subfraksi, serta mengidentifikasi senyawa pada subfraksi daun Ficus adenosperma Warb. yang memiliki aktivitas antioksidan berdasarkan hasil uji DPPH. Ekstraksi dilakukan dengan metode sonikasi, dilanjutkan ekstraksi cair-cair menggunakan pelarut air, etil asetat, dan n-heksana. Identifikasi subfraksi dilakukan dengan menggunakan metode FTIR dan HPLC. Hasil uji kandungan senyawa fenolik dan flavonoid menunjukkan bahwa ekstrak etanol daun Ficus adenosperma Warb. memiliki kadar total fenolik sebesar 81,122 mg GAE/gr dan kadar total flavonoid sebesar 57,984 mg QE/gr. Ketiga fraksi yang dihasilkan dari ekstraksi cair-cair menunjukkan aktivitas antioksidan, dengan nilai persen inhibisi fraksi etil asetat sebesar 89,45%, fraksi air sebesar 58,62%, sedangkan fraksi n-heksana menunjukkan persen inhibisi sebesar 21,13%. Subfraksi aktif antioksidan yang berhasil dipisahkan berasal dari fraksi air yang ditandai dengan terbentuknya kristal. Senyawa aktif antioksidan yang teridentifikasi kemungkinan besar adalah kuersetin. / Free radicals are foreign substances that can infiltrate the human body and potentially damage the immune system. Antioxidants are compounds that can inhibit or prevent the formation of free radicals. Naturally, the body has antioxidants, but their quantity is often insufficient to prevent an excess of free radicals. Therefore, exogenous antioxidants are needed. One plant that contains natural antioxidants is the genus Ficus. This study aims to determine the fractions, sub-fractions, and identify compounds in the sub-fractions of Ficus adenosperma Warb. leaves with antioxidant activity based on the DPPH assay results. Extraction was carried out using the sonication method, followed by liquid-liquid extraction using water, ethyl acetate, and n-hexane solvents. Sub-fraction identification was done using FTIR and HPLC methods. The results of phenolic and flavonoid content tests showed that the ethanol extract of Ficus adenosperma Warb. leaves has a total phenolic content of 81.122 mg GAE/g and a total flavonoid content of 57.984 mg QE/g. The three fractions obtained from liquid-liquid extraction showed antioxidant activity, with the inhibition percentage of the ethyl acetate fraction at 89.45%, the water fraction at 58.62%, and the n-hexane fraction at 21.13%. The active antioxidant sub-fraction separated from the water fraction was marked by crystal formation. The active antioxidant compound identified is most likely quercetin.
