Abstract
Jerawat merupakan kondisi inflamasi kulit yang umum terjadi pada remaja dan dewasa muda. Pengobatan topikal yang sering digunakan menimbulkan efek samping seperti iritasi dan fotosensitivitas. Hal ini mendorong pengembangan alternatif yang lebih aman, seperti pemanfaatan tanaman herbal. Cyperus rotundus (rumput teki), tanaman yang banyak ditemukan di Indonesia, diketahui memiliki aktivitas antibakteri. Ekstrak etanol umbi rumput teki memiliki potensi menghambat pertumbuhan Staphylococcus epidermidis dengan MIC 250 μg/ml. Formulasi nanospray gel sebagai sistem penghantaran topikal berbasis nanoteknologi diharapkan dapat meningkatkan efektivitas dan penetrasi zat aktif. Penelitian ini bertujuan untuk menguji aktivitas antibakteri ekstrak etanol C. rotundus terhadap S. epidermidis dan C. acnes, memformulasi nanospray gel, serta menentukan konsentrasi efektif untuk aktivitas antibakteri optimal. Metode yang digunakan meliputi ekstraksi sampel dengan metode maserasi, pengujian MIC ekstrak etanol C. rotundus, formulasi sediaan nanospray gel, uji aktivitas antibakteri sediaan nanospray gel menggunakan metode disk diffusion, serta uji karakteristik fisiko-kimia sediaan nanospray gel. Diperoleh hasil rendemen sebesar 8% menggunakan metode ekstraksi maserasi. Ekstrak umbi rumput teki menghasilkan nilai MIC 350.000 μg/ml. Didapatkan bahwa FI (ekstrak 1 gram) memenuhi sebagian besar syarat evaluasi fisiko-kimia sediaan. Pengujian antimikroba sediaan menunjukkan FI menghasilkan aktivitas antimikroba terbaik untuk S. epidermidis, dan FII menghasilkan aktivitas antimikroba terbaik untuk C. acnes./Acne is an inflammatory skin condition commonly found in teenagers and young adults. Commonly used topical treatments can cause side effects such as irritation and photosensitivity. This has encouraged the development of safer alternatives, such as the use of herbal plants. Cyperus rotundus (nutgrass), a plant commonly found in Indonesia, is known to possess antibacterial activity. Ethanol extract of C. rotundus tubers has shown potential in inhibiting the growth of Staphylococcus epidermidis with a Minimum Inhibitory Concentration (MIC) of 250 μg/ml. The formulation of a nanospray gel as a nanotechnology-based topical drug delivery system is expected to improve the effectiveness and penetration of the active compound. This study aims to evaluate the antibacterial activity of C. rotundus ethanol extract against S. epidermidis and C. acnes, formulate a nanospray gel, and determine the effective concentration for optimal antibacterial activity. The methods are sample extraction by maceration method, MIC testing of C. rotundus ethanol extract, formulation of nanospray gel preparation, antibacterial activity test of nanospray gel using disk diffusion method, and physico-chemical characteristics test of nanospray gel preparation. An 8% yield was obtained through maceration extraction. The tuber extract had an MIC of 350,000 μg/ml. Formula I (1 gram extract) met most physicochemical evaluation criteria. Antimicrobial testing showed that FI was most effective against S. epidermidis, while FII showed the best activity against C. acnes.
