Abstract
Radikal bebas dapat menyebabkan stres oksidatif yang dapat menyebabkan penyakit degeneratif yang berbahaya bagi kesehatan, untuk mencegah radikal bebas dapat menggunakan antioksidan. Pada penelitian ini menggunakan tanaman milk thistle sebagai sumber antioksidan alami.Tanaman milk thistle (Silybum marianum L.) diketahui sebagai tanaman obat dan sudah digunakan dalam pengobatan selama lebih dari 2000 tahun. Salah satu khasiat farmakologis nya yaitu antioksidan. Diketahui aktivitas antioksidan terdapat pada bagian biji tanaman yang memiliki kandungan komponen utama yaitu silymarin sebesar 70%-80%. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan identifikasi senyawa antioksidan dari biji tanaman milk thistle (Silybum marianum L.). Penelitian ini
menggunakan metode maserasi untuk mengekstrak biji tanaman dan menggunakan metode DPPH untuk uji aktivitas antioksidan dan pengukuran dengan spektrofotometer Uv-Vis. Isolasi senyawa menggunakan metode fraksinasi asam basa dan kromatografi kolom. Selanjutnya, Identifikasi senyawa menggunakan LC
MS/MS. Didapatkan hasil total fenolik ekstrak metanol dan fraksi asam biji milk thistle yaitu 82,75 mg GAE/ g fraksi dan 621,03 mg GAE/g ekstrak. Total flavonoid ekstrak metanol dan fraksi asam biji milk thistle yaitu 11,21 mg QE/g ekstrak dan 17,02 mg QE/g fraksi. Nilai penghambatan radikal bebas (IC50) pada ekstrak dan fraksi asam dengan konsentrasi 500 ppm yaitu 435,16 ppm dan 161.31 ppm. Hasil LC-MS/MS menunjukan bahwa fraksi asam memiliki senyawa silychristin, silydianin, dan silybin A dan B. / Free radicals can cause oxidative stress that can cause degenerative diseases that are harmful to health, to prevent free radicals can use antioxidants. Milk thistle plants (Silybum marianum L.) are known as medicinal plants and have been used in medicine for more than 2000 years. One of its pharmacological properties is antioxidants. It is known that antioxidant activity is found in the seeds of plants that have a main component content of silymarin of 70% -80%. This study aims to determine the antioxidant activity and identification of antioxidant compounds from milk thistle seeds (Silybum marianum L.). The total phenolic results of the methanol extract and the acid fraction of milk thistle seeds were 82.75 mg GAE/g fraction and 621.03 mg GAE/g extract. The total flavonoids of the methanol extract and the acid fraction of milk thistle seeds were 11.21 mg QE/g extract and 17.02 mg QE/g fraction. The free radical inhibition value (IC50) in the extract and acid fraction with a concentration of 500 ppm was 435.16 ppm and 161.31 ppm. The LC-MS/MS results showed that the acid fraction contained silychristin, silydianin, and silybin A and B compounds.
