Abstract
Pengobatan tradisional berbasis tanaman obat diminati masyarakat karena memiliki efek samping yang rendah, biaya terjangkau, dan kemudahan penggunaannya. Salah satu tanaman potensial sebagai pengobatan tradisional adalah kulit batang biwa yang telah diteliti memiliki aktivitas sebagai antifungi terhadap Cryptococcus neoformans, antibakteri terhadap Propionibacterium acnes dan Staphylococcus aureus. Penelitian ini bertujuan untuk mengkaji toksisitas akut ekstrak etanol 96% kulit batang biwa menggunakan Acute Toxic Class sesuai pedoman OECD 423, menganalisis efek toksisitas terhadap organ hati dan ginjal melalui identifikasi hitopatologi, mengevaluasi pemberian dosis aman ekstrak satu kali sehari selama 14 hari terhadap profil hematologi dan biokimia darah (SGOT, SGPT, kreatinin) tikus jantan wistar. Hasil penelitian didapatkan ekstrak etanol 96% kulit batang biwa berdasarkan klasifikasi GHS (Global Harmonized System) cut-off LD50 kategori 5 (tidak terklasifikasikan). Pengujian harian menggunakan ekstrak dosis aman 2000 mg/kg satu kali sehari selama 14 hari menghasilkan perbedaan kadar SGPT yang signifikan mempengaruhi organ hati. Selain itu terdapat perbedaan signifikan kadar kreatinin hari ke-14 mengindikasikan ekstrak etanol 96% kulit batang biwa mempengaruhi kerusakan ginjal. Evaluasi hematologi terdapat perbedaan signifikan jumlah retikulosit disebabkan ekstrak kulit batang biwa mempengaruhi sistem hematopoietik. / Herbal-based traditional medicine is widely favored by the public due to its low
side effects, affordability, and ease of use. One promising medicinal plant is loquat (Eriobotrya japonica) stem bark, which has shown antifungal activity against
Cryptococcus neoformans and antibacterial effects against Propionibacterium acnes and Staphylococcus aureus. This study aims to evaluate the acute toxicity of
the 96% ethanol extract of loquat stem bark using the Acute Toxic Class (ATC) method following OECD guideline 423. It also aims to analyze its toxic effects on the liver and kidneys through histopathological examination, and to evaluate the effects of a safe dose administered once daily for 14 days on hematological and blood biochemical parameters, including SGOT, SGPT, and creatinine, in male Wistar rats. The results showed that the extract falls under GHS (Globally Harmonized System) toxicity category 5 or is unclassified, indicating low acute
toxicity. Administration of a 2000 mg/kg dose daily for 14 days led to a significant reduction in SGPT levels on day 14, a significant increase in creatinine levels indicated potential kidney damage. In addition, hematological examination showed significant changes in the number of reticulocytes indicating that the extract can affect the hematopoietic system.
