Abstract

Cabai katokkon (Capsicum chinense Jacq.) dari Toraja, Sulawesi Selatan, dikenal memiliki kandungan capsaicin tinggi, yang memberikan rasa pedas khas. Capsaicin, komponen utama yang memberikan rasa pedas, memiliki berbagai manfaat farmakologis, termasuk sifat analgesik, antioksidan, antimikroba, dan pencegahan kanker. Oleh karena itu, diperlukan pengembangan metode ekstraksi yang optimal untuk meningkatkan rendemen capsaicin secara efisien dan terarah, guna mendukung aplikasinya dalam berbagai bidang. Penelitian ini mengoptimalkan metode Ultrasound-Assisted Extraction (UAE) menggunakan Response Surface Methodology (RSM) dengan Box-Behnken Design (BBD) untuk meningkatkan rendemen capsaicin. Tiga variabel utama yang diuji adalah waktu ekstraksi, indeks polaritas pelarut, dan rasio pelarut terhadap bahan. Analisis secara kualitatif dilakukan dengan Kromatografi Lapis Tipis (KLT) dan analisis kuantitatif dilakukan dengan High Performance Liquid Chromatography (HPLC). Hasil menunjukkan bahwa indeks polaritas pelarut dan interaksi waktu ekstraksi dan indeks polaritas berpengaruh signifikan. Prediksi kondisi optimal dalam rentang variabel yang diuji diperoleh pada ekstraksi 9 menit, indeks polaritas 4,4 (etil asetat), dan rasio pelarut:bahan 1:10, menghasilkan 44,69 mg capsaicin per 500 mg ekstrak. Namun, diprediksi masih terus meningkat pada waktu ekstraksi semakin lama, indeks polaritas lebih non polar dan rasio pelarut yang lebih banyak. Hasil diperoleh dari persamaan model polinomial kuadratik yang dikurangi dari sembilan variabel menjadi tujuh variabel./Katokkon chili (Capsicum chinense Jacq.) from Toraja, South Sulawesi, is known to have a high capsaicin content, which gives it a distinctive spicy taste. Capsaicin, the main component that gives it a spicy taste, has various pharmacological benefits, including analgesic, antioxidant, antimicrobial, and cancer prevention properties. Therefore, it is necessary to develop an optimal extraction method to increase capsaicin yield efficiently and in a targeted manner, to support its application in various fields. This study optimized the Ultrasound-Assisted Extraction (UAE) method using Response Surface Methodology (RSM) with Box-Behnken Design (BBD) to increase capsaicin yield. The three main variables tested were extraction time, solvent polarity index, and solvent to material ratio. Qualitative analysis was carried out using Thin Layer Chromatography (TLC) and quantitative analysis was carried out using High Performance Liquid Chromatography (HPLC). The results showed that the solvent polarity index and the interaction of extraction time and polarity index had a significant effect. The prediction of optimal conditions in the range of variables tested was obtained at 9 minutes of extraction, a polarity index of 4.4 (ethyl acetate), and a solvent:material ratio of 1:10, producing 44.69 mg of capsaicin per 500 mg of extract. However, it is predicted to continue to increase at longer extraction times, more non-polar polarity indices and higher solvent ratios. The results were obtained from the quadratic polynomial model equation which was reduced from nine variables to seven variables.
