Abstract
Inflamasi adalah respons biologis tubuh terhadap cedera, infeksi, atau paparan rangsangan yang melibatkan sistem pertahanan tubuh. Rimpang kunyit (Curcuma longa Linn.) mengandung senyawa aktif kurkumin yang memiliki aktivitas antiinflamasi, namun termasuk dalam BCS Kelas II. Oleh karena itu, formulasi Nanostructured Lipid Carriers (NLC) dilakukan untuk meningkatkan bioavailabilitas kurkumin. Penelitian ini memformulasikan ekstrak rimpang kunyit 5% menjadi NLC yang dimuat dalam krim dengan variasi 5%, 25%, dan 50%. Ekstraksi dilakukan menggunakan metode sonikasi dengan hasil rendemen 26,2933%. Metode pembuatan NLC melalui metode emulsifikasi dengan komponen lipid padat, lipid cair, surfaktan, dan ko-surfaktan. Karakterisasi NLC meliputi uji organoleptis, viskositas, dan ukuran partikel yang memenuhi syarat, namun indeks polidispersitas dan zeta potensial belum memenuhi persyaratan. Evaluasi fisik krim NLC, seperti pH, tipe krim, homogenitas, daya lekat, dan daya sebar, serta uji stabilitas menunjukkan hasil yang baik. Hasil uji antiinflamasi menunjukkan bahwa krim NLC ekstrak rimpang kunyit konsentrasi 50% (FIII) memiliki %DAI sebesar 88,65%, yang lebih tinggi dibandingkan dengan krim ekstrak rimpang kunyit. Oleh karena itu, formulasi krim NLC terbukti mampu meningkatkan efektivitas antiinflamasi ekstrak rimpang kunyit pada tikus putih galur Wistar yang diinduksi karagenan 1%.

Inflammation is the body's biological response to injury, infection, or exposure to stimuli involving the body's defense system. Turmeric rhizome (Curcuma longa Linn.) contains the active compound curcumin, which exhibits anti-inflammatory activity but belongs to BCS Class II. Therefore, the formulation of Nanostructured Lipid Carriers (NLC) was conducted to enhance curcumin's bioavailability. This study formulated 5% turmeric rhizome extract into NLC-loaded cream with variations of 5%, 25%, and 50%. Extraction was performed using the sonication method, yielding 26.2933%. The NLC was prepared using the emulsification method with solid lipids, liquid lipids, surfactants, and co-surfactants. NLC characterization included organoleptic testing, viscosity, and particle size, which met the required criteria, although the polydispersity index and zeta potential did not meet the standards. The physical evaluation of NLC cream, including pH, cream type, homogeneity, adhesion, spreadability, and stability tests, showed favorable results. The anti-inflammatory test results demonstrated that the NLC cream containing 50% turmeric rhizome extract (FIII) had a %DAI of 88.65%, which was higher than the turmeric rhizome extract cream. Therefore, the NLC cream formulation effectively enhanced the anti-inflammatory efficacy of turmeric rhizome extract in Wistar rats induced with 1% carrageenan.
