Abstract

Cabai Hiyung merupakan varietas dari tanaman cabai rawit yang berasal dari Kalimatan Selatan. Tanaman ini memiliki kandungan senyawa capsaicin 17 kali yang lebih besar dibandingkan dengan cabai rawit biasa. Maka dari itu, cabai hiyung memiliki potensi untuk memberikan hasil ekstraksi capsaicin dalam jumlah banyak. Proses ekstraksi capsaicin menggunakan metode Ultrasonic Assisted Extraction (UAE) tidak terlepas dari pertimbangan parameter selama ekstraksi berlangsung agar memperoleh hasil ekstraksi capsaicin yang maksimal. Pertimbangannya meliputi jenis pelarut, durasi ekstraksi, dan rasio antara bahan dengan pelarut. Walaupun pertimbangan terhadap parameter telah dilakukan, tetapi tidak semua proses ekstraksi berlangsung secara optimal, maka untuk mengatasi kondisi tersebut dapat melakukan optimasi menggunakan program Response Surface Methodology (RSM) dengan memakai parameter proses ekstraksi sebagai variabel yang akan dimasukan ke dalam kinerja program tersebut. Penelitian ini juga melakukan pengujian secara kualitatif dalam mengidentifikasi keberadaan capsaicin denan metode kromatografi lapis tipis dan kuantitatif dengan memakai alat Ultra High Performance Liquid Chromatography (UHPLC) dalam menentukan kadar capsaicin pada sampel. Kondisi maksimum didapatkan dengan meningkatkan kenaikan nilai yield dengan menggunakan arah gradien sejauh feasible dari hasil optimasi dengan pendekatan RSM-BBD adalah durasi 6 menit, pelarut etil asetat (4,4) , dan rasio bahan antara pelarut adalah 1:4,6164 atau 23 mL memperoleh yield sampel sebesar 10,3223 ppm dalam larutan sampel 500 ppm. Hasil tersebut berdasarkan pengurangan jumlah variabel yang digunakan dari 9 variabel menjadi 5 variabel pada model polinominal kuadratik./The Hiyung chili is a variety of chili plants originating from South Kalimantan. This plant has a capsaicin compound content 17 times greater than ordinary cayenne pepper. Therefore, Hiyung chili has the potential to provide more capsaicin extraction. The extraction process of capsaicin using Ultrasonic Assisted Extraction (UAE) is inseparable from the consideration of various parameters during extraction to achieve optimal yields. These include the type of solvent, extraction duration, and the ratio of raw material to solvent. However, even with these parameters, not all extraction processes are optimal. Optimization can be conducted using Response Surface Methodology (RSM), with the extraction process parameters serving as variables for the program’s performance. This research also conducts qualitative tests to identify capsaicin using thin-layer chromatography and quantitative analysis via Ultra High-Performance Liquid Chromatography (UHPLC) to determine capsaicin sample levels. The maximum condition is obtained by increasing the yield value increase by using the gradient direction as far as feasible from the optimization results with the RSM-BBD approach are a duration of 6 minutes, ethyl acetate solvent (4,4), and the ratio of materials between solvents is 1:4.6164 or 23 mL obtaining a sample yield of 10.3223 ppm in a sample solution of 500 ppm. These results are based on the reduction of the number of variables used from 9 variables to 5 variables in the quadratic polynomial model.
