Abstract
Ampas kopi (Coffea arabica L.) bermanfaat sebagai agen eksfoliasi kulit bibir, sedangkan kulit manggis (Garcinia mangostana L.) memiliki efek antioksidan untuk permasalahan hiperpigmentasi pada bibir. Tujuan penelitian yakni membuat sediaan lip scrub emulgel dari kombinasi keduanya dan memenuhi evaluasi fisik, mengetahui nilai aktivitas antioksidan dengan metode DPPH serta uji keamanan yang meliputi uji iritasi dan uji cemaran mikroba (Angka Lempeng Total dan Angka Kapang Khamir). Hasil menunjukkan ekstrak etanol 96% kulit manggis mengandung polifenol yang memiliki aktivitas antioksidan utama. Sediaan lip scrub emulgel diformulasikan menjadi tiga konsentrasi ampas kopi yang berbeda yakni 2%;4%;6%. Hasil evaluasi fisik meliputi uji pH, homogenitas, daya sebar, daya lekat dan cycling test telah memenuhi persyaratan. Selanjutnya, dilakukan pengujian hedonik dengan hasil terpilih sediaan F2;4% yang paling disukai 10 responden. Sediaan lip scrub emulgel memiliki aktivitas antioksidan dengan nilai 〖IC〗_50 sebesar 14.240,86 μg/ml. Hasil ALT telah memenuhi syarat yakni sebesar <〖10〗^1 koloni/ml. Namun, hasil AKK belum memenuhi syarat yakni sebesar 〖8,4×10〗^5 koloni/ml. Uji iritasi sediaan lip scrub emulgel dilakukan terhadap 10 sukarelawan dengan hasil skor 0,0 atau tidak mengiritasi./ Coffee grounds (Coffea arabica L.) serve as an exfoliating agent for lips, while mangosteen peel (Garcinia mangostana L.) provides antioxidant effects to address lip hyperpigmentation. This study aimed to develop a lip scrub emulgel combining both ingredients, evaluate its physical properties, determine antioxidant activity using the DPPH method, and assess its safety through irritation and microbial contamination tests (Total Plate Count and Yeast Mold Count). Ethanol extract (96%) of mangosteen peel was found to contain polyphenols as the main antioxidant component. The emulgel was formulated with three different concentrations of coffee grounds: 2%, 4%, and 6%. Evaluation showed that all physical parameters—pH, homogeneity, spreadability, adhesion, and cycling test—met acceptable standards. Hedonic testing revealed that Formula 2 (4%) was the most preferred by 10 respondents. The formulation showed antioxidant activity with an 〖IC〗_50 value of 14,240.86 μg/mL. The Total Plate Count met the requirement (<10¹ colonies/mL), but the Yeast Mold Count did not, with 8.4×10⁵ colonies/mL. Irritation testing on 10 volunteers showed no signs of irritation (score 0.0).
