Abstract
Pertumbuhan bakteri S. aureus dan C. acnes merupakan salah satu penyebab terjadinya jerawat. Untuk mengatasi permasalahan resistensi serta efek samping yang disebabkan oleh obat jerawat (antibiotik), dilakukan formulasi liposom biji pinang (Areca catechu L.) yang mengandung senyawa antibakteri. Biji Pinang dimaserasi dengan pelarut etanol 96% dan dibentuk tiga formulasi liposom dengan variasi ekstrak 2,5, 5,5, dan 7,5%. Ekstrak dan liposom biji pinang memiliki aktivitas antibakteri terhadap bakteri S. aureus dan C. acnes yang tidak berbeda signifikan. Ekstrak biji pinang memiliki nilai KHM terhadap S. aureus pada konsentrasi 781,25 ppm dan bakteri C. acnes pada konsentrasi 1562,5 ppm. Nilai KBM yang dihasilkan ekstrak biji pinang terhadap bakteri S. aureus dan C. acnes adalah 6250 ppm. Pada bakteri S. aureus dan C. acnes, liposom biji pinang memberikan nilai KHM 3.125 ppm dan nilai KBM 12.500 ppm. Hasil uji ukuran vesikel (418,3-464,4 nm) dan uji efisiensi penjerapan (84-87%) pada liposom memenuhi persyaratan karakterisasi liposom. Nilai zeta potensial (-9,2 sampai -7,4 mV) dan indeks polidispersitas (1,841-2,618) menunjukkan kecenderungan liposom untuk membentuk agregat dan ukuran vesikel yang tidak homogen. Ketiga formulasi liposom menunjukkan kemampuan untuk menjaga kestabilan senyawa aktif yang cukup baik dengan hasil efisiensi penjerapan diatas 80% setelah dilakukan cycling test. / The growth of Staphylococcus aureus and Cutibacterium acnes is one of the primary causes of acne. To address issues of resistance and the side effects associated with acne treatments (antibiotics), a liposomal formulation containing antibacterial compounds from areca nut (Areca catechu L.) was developed. The areca nut was macerated using 96% ethanol as the solvent, and three liposomal formulations were prepared with extract concentrations of 2,5%, 5,5%, and 7,5%. Both the extract and the liposomal formulations exhibited antibacterial activity against S. aureus and C. acnes, with no significant difference between them. The areca nut extract showed a Minimum Inhibitory Concentration (MIC) of 781.25
ppm against S. aureus and 1562.5 ppm against C. acnes. The Minimum Bactericidal Concentration (MBC) of the extract for both bacteria was 6250 ppm. For the liposomal formulations, the MIC and MBC values against both S. aureus and C. acnes were 3125 ppm and 12,500 ppm, respectively. The vesicle size (418.3–464.4 nm) and entrapment efficiency (84–87%) of the liposomes met the characterization requirements. However, the zeta potential values (−9.2 mV to −7.4 mV) and polydispersity index (1.841–2.618) indicated a tendency for vesicle aggregation and non-uniform size distribution. Despite this, all three liposomal formulations demonstrated good ability to maintain the stability of the active compounds, with entrapment efficiency remaining above 80% after cycling tests.
