Abstract

Stress oksidatif merupakan suatu kondisi yang dapat menyebabkan terjadinya penyakit yang bersifat kronis seperti kanker, jantung, hingga menurunnya sistem kekebalan tubuh. Oleh karena hal ini, diperlukan keberadaan antioksidan untuk mencegah terjadinya kondisi seperti ini. Salah satu tanaman yang memiliki kandungan antioksidan adalah tanaman kayu ules (Helicteres isora) terkhusus pada bagian buahnya, diketahui mengandung senyawa golongan fenolik dan flavonoid. Tujuan penelitian ini adalah untuk mengetahui aktivitas antioksidan dengan menggunakan parameter IC_50 serta mengidentifikasi kandungan senyawa aktif dalam fraksi asam buah tanaman kayu ules. Didapatkan hasil uji total fenolik pada fraksi asam buah tanaman kayu ules sebesar 103,365 ± 0,775 mgGA/g dan uji total flavonoid sebesar dan 97,884 ± 0,207 mgQE/g. Kemudian untuk mengukur aktivitas antioksdian digunakan metode DPPH dan didapatkan hasil sebesar 816,606 µg/ml. Senyawa fraksi asam tanaman ini, selanjutnya dimurnikan dengan menggunakan kolom kromatografi dan didaptkan satu fraksi utama (fraksi 6) untuk dimurnikan dengan menggunakan LC-MS/MS. Data LC-MS/MS diduga menunjukkan adanya kandungan senyawa kaempferol pada fraksi asam yang diuji. / Oxidative stress is a condition that can cause chronic diseases such as cancer, heart disease, and decreased immune system. Therefore, antioxidants are needed to prevent such conditions. One of the plants that has antioxidant content is the ules wood plant (Helicteres isora) especially in the fruit, known to contain phenolic and flavonoid compounds. The purpose of this study was to determine antioxidant activity using the IC_50 parameter and to identify the content of active compounds in the fruit acid fraction of the ules wood plant. The results of the total phenolic test in the fruit acid fraction of the ules wood plant were 103.365 ± 0.775 mgGA / g and the total flavonoid test was 97.884 ± 0.207 mgQE / g. Then to measure antioxidant activity, the DPPH method was used and the results were 816.606 µg / ml. The compound of this plant acid fraction was then purified using column chromatography and one main fraction (fraction 6) was obtained to be purified using LC-MS/MS. LC-MS/MS data is suspected to indicate the presence of kaempferol compounds in the tested acid fraction.
