Abstract

Radikal bebas dapat menyebabkan kerusakan pada bibir, seperti bibir kering dan pecah-pecah. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan ekstrak etanol 96% daun bayam merah (Amaranthus tricolor L.) serta aplikasinya dalam sediaan lip balm. Ekstrak diperoleh melalui metode Ultrasound-Assisted Extraction (UAE) menggunakan pelarut etanol 96%, kemudian diformulasikan menjadi sediaan lip balm dengan konsentrasi ekstrak 3%, 5%, dan 7%. Uji aktivitas antioksidan dilakukan menggunakan metode DPPH untuk menentukan nilai IC50 pada ekstrak dan sediaan lip balm. Hasil penelitian menunjukkan bahwa ekstrak etanol 96% daun bayam merah memiliki aktivitas antioksidan yang sangat kuat dengan nilai IC50 sebesar 46,054±0,560 ppm. Sementara itu, sediaan lip balm menunjukkan aktivitas antioksidan yang tergolong sangat lemah, dengan nilai IC50 berturut-turut: F1 (3%) sebesar 3740,39±65,13 ppm, F2 (5%) sebesar 3485,69±31,91 ppm, dan F3 (7%) sebesar 3088,82±11,95 ppm. Formula F3 yang mengandung 7% ekstrak dengan IC50 terendah, merupakan formula paling optimal yang didasarkan pada analisis statistik./ Free radicals can cause damage to the lips, leading to dry and chapped lips. This study aims to determine the antioxidant activity of 96% ethanol extract of red spinach leaves (Amaranthus tricolor L.) and its application in lip balm formulations. The extract was obtained through the Ultrasound-Assisted Extraction (UAE) method using 96% ethanol as the solvent, and then formulated into lip balm preparations with extract concentrations of 3%, 5%, and 7%. The antioxidant activity was tested using the DPPH method to determine the IC50 value for both the extract and the lip balm formulations. The results showed that the 96% ethanol extract of red spinach leaves exhibited very strong antioxidant activity with an IC50 value of 46.054 ppm. Meanwhile, the lip balm formulations demonstrated very weak antioxidant activity, with IC50 values of F1 (3%) at 3740,39 ppm, F2 (5%) at 3485,69 ppm, and F3 (7%) at 3088,82 ppm, respectively. Formula F3, containing 7% extract with the lowest IC50, is considered the most optimal formulation based on statistical analysis.
