Abstract
Kejadian diabetes di Indonesia diprediksi akan meningkat pesat, dari 19,5 juta orang terdiagnosis pada tahun 2021 menjadi 28,6 juta pada tahun 2045. Efek samping dari obat-obatan sintesis, menjadi dasar penggunaan bahan yang berasal dari tumbuhan herbal sebagai alternatif dalam mengontrol kadar gula darah seperti daun ketul. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas dan dosis efektif ekstrak etanol 70% daun ketul sebagai antihiperglikemik pada hewan uji mencit yang diinduksi aloksan. Daun Ketul memiliki potensi penurunan kadar glukosa melalui senyawa fenol dan flavonoid. Pengujian kadar flavonoid dan fenol total dilakukan dengan metode kolorimetri. Pengujian efek antihiperglikemik dilakukan dengan metode induksi aloksan. Analisis data dilakukan dengan menggunakan uji normalitas, homogenitas, kruskal-wallis, dan man whitney. Hasil pengujian menunjukkan kadar kuersetin, rutin, dan asam galat sebesar 14,067 ± 2,64 mgQE/g ekstrak; 22,936 ± 3,77 mgRE/g ekstrak; 25,205 ± 1,61 mgGAE/g ekstrak. Uji antihiperglikemik menunjukkan persentase penurunan sebesar 48,00%; 68,08%; dan 81,98%. Uji kruskal wallis menunjukkan nilai signifikansi 0,005 (<0,05) dan nilai signifikansi dosis terkecil adalah 0,465 (p≥0,05) terhadap kontrol positif. Analisis pengaruh ekstrak terhadap berat badan menunjukkan signifikansi sebesar 0,159 (p≥0,05). Hasil penelitian menunjukkan bahwa ekstrak etanol 70% daun ketul memiliki aktivitas anthiperglikemik dengan dosis efektif adalah 100 mg/kgBB dan tidak mempengaruhi berat badan hewan uji / The incidence of diabetes in Indonesia is predicted to increase rapidly, from 19.5 million people in 2021 to 28.6 million in 2045. Side effects of synthetic drugs are the basis for the use of ingredients derived from herbal plants as an alternative in controlling blood sugar levels such as ketul leaves. The purpose of this study was to determine the activity and effective dose of ethanol 70% extract of ketul leaves as an antihyperglycemic in alloxane-induced mice test animals. Ketul leaves have the potential to reduce glucose levels through phenol and flavonoid compounds. Testing of total flavonoid and phenol levels was carried out by colorimetric method. Testing of the antihyperglycemic effect was carried out by the alloxane induction method. Data analysis was carried out using normality, homogeneity, kruskal-wallis, and man whitney tests. The test results showed levels quarcetine, rutine, and gallic acid of 14.067 ± 2.64 mgQE/g extract; 22.936 ± 3.77 mgRE/g extract; 25.205 ± 1.61 mgGAE/g extract. The antihyperglycemic test showed a percentage decrease of 48.00%; 68.08%; and 81.98%. The kruskal wallis test showed a significance value of 0.005 (<0.05) and the smallest dose significance value was 0.465 (p≥0.05) against positive controls. Analysis of the effect of the extract on body weight showed a significance of 0.159 (p≥0.05). The results showed that ethanol 70% extract ketul leaves had antihyperglycemic activity with an effective dose of 100 mg/BW and did not affect the body weight of the test animals.
