Abstract

Diabetes merupakan suatu penyakit metabolik kronis yang disebabkan ketika pankreas tidak dapat memproduksi insulin dalam jumlah yang cukup atau ketika tubuh sudah tidak dapat menggunakan insulin secara efektif, sehingga mejadi penyebab penyakit diabetes melitus. Word Health Organization (WHO) memperkirakan jumlah penderita DM tipe 2 di Indonesia akan meningkat signifikan hingga 21,3 juta jiwa pada tahun 2030 mendatang. Kandungan metabolit sekunder pada tanaman berupa flavonoid, tannin, saponin, dapat menjadi agen antihiperglikemik. Oleh karena itu, penelitian ini dilakukan untuk mengetahui dan menganalisis aktivitas ekstrak etanol 96% batang biwa (Eriobotrya japonica Caulis (Thunb.) Lindl.). Metode penelitian dilakukan secara eksperimental di laboratorium. Ekstrak diperoleh dengan metode maserasi menggunakan pelarut etanol 96%, dilanjutkan dengan skrining fitokimia dan Uji kuantitatif flavonol dan fenolik total. Uji antihiperglikemik dilakukan dengan cara menginduksi mencit dengan aloksan. Hasil uji skrining positif senyawa alkaloid, flavonoid, saponin, tanin, fenolik, dan steroid. Pada uji kuantitatif senyawa flavonol dan tanin total mengandung 30,583±0,769 mgRE/g dan 459,539±9,11 mgTAE/g ekstrak. Pada uji antihiperglikemik, esktrak etanol 96% batang biwa memiliki aktivitas menurunkan kadar glukosa darah mencit, dimana kontrol positif (61,97%) dan kelompok ekstrak dosis 100 mg/kgBB, 300 mg/kgBB dan 500 mg/kgBB (26,90%, 31,69% dan 52,29%), sehingga dosis efektif ekstrak adalah 500 mg/kgBB mencit./ Diabetes is a chronic metabolic disease caused when the pancreas cannot produce enough insulin or when the body cannot use insulin effectively, thus causing diabetes mellitus. The World Health Organization (WHO) estimates that the number of type 2 diabetes patients in Indonesia will increase significantly to 21.3 million by 2030. Secondary metabolites in plants, such as flavonoids, tannins, and saponins, can act as antihyperglycemic agents. Therefore, this study was conducted to investigate and analyze the activity of 96% ethanol extract from the stems of the loquat tree (Eriobotrya japonica Caulis (Thunb.) Lindl.). The research was conducted experimentally in a laboratory. The extract was obtained using the maceration method with 96% ethanol as the solvent, followed by phytochemical screening and quantitative testing of flavonols and total phenolics. The antihyperglycemic test was conducted by inducing mice with alloxan. The screening test results were positive for alkaloids, flavonoids, saponins, tannins, phenolics, and steroids. In the quantitative test, the flavonol and total tannin content was 30.583±0.769 mgRE/g and 459.539±9.11 mgTAE/g of extract. In the antihyperglycemic test, the 96% ethanol extract of biwa stem showed activity in lowering blood glucose levels in mice, with the positive control (61.97%) and the extract groups at doses of 100 mg/kgBW, 300 mg/kgBW, and 500 mg/kgBW (26.90%, 31.69%, and 52.29%), respectively. Thus, the effective dose of the extract is 500 mg/kgBW for mice.
