Abstract
Tanaman kecipir (Psophocarpus tetragonolobus (L.) DC.) merupakan tanaman kacang-kacangan dengan sumber antioksidan, terutama senyawa polifenol dan flavonoid yang berpotensi melindungi dari stres oksidatif dan menurunkan kadar gula darah. Flavonoid mampu melindungi dari stres oksidatif dan mengurangi ROS pada diabetes maupun hiperglikemik. Pelarut etanol 70% dipilih karena kemampuannya menarik senyawa yang bersifat polar dan dapat menghasilkan beragam kelompok senyawa metabolit sekunder. Penelitian ini bertujuan menganalisis kandungan metabolit sekunder dari ekstrak etanol 70% buah kecipir dan menganalisis aktivitas antihiperglikemik pada mencit putih jantan yang diinduksi aloksan. Uji dilakukan terhadap lima kelompok: kontrol negatif Na CMC 0,5%; kontrol positif glibenklamid 0,0208 mg/32gBB; dosis ekstrak kecipir 100 mg/kgBB; 300 mg/kgBB; dan 500 mg/kgBB. Kadar glukosa darah dianalisis dengan metode GOD-PAP menggunakan fotometer. Uji skrining fitokimia menunjukkan kandungan senyawa alkaloid, flavonoid, tanin, dan fenolik. Uji antihiperglikemik menunjukkan ekstrak mampu menurunkan kadar glukosa darah dengan persentase penurunan berturut-turut adalah 60,39%; 64,88%; dan 69,71% pada tiga dosis tersebut. Uji statistik SPSS menunjukkan terdistribusi normal (p=0,457). Uji One Way ANOVA menunjukkan hasil perbedaan signifikan (p=0,001), dan uji Post Hoc Tamhane menunjukkan tidak ada perbedaan bermakna antara kontrol positif dan ketiga dosis ekstrak. Hasil yang diperoleh dosis 100 mg/kgBB merupakan dosis efektif yang sebanding dengan kontrol positif glibenklamid. / The winged bean (Psophocarpus tetragonolobus (L.) DC.) is leguminous plant rich in antioxidants, particularly polyphenols and flavonoids which have potential protective effects against oxidative stress and blood glucose elevation. Flavonoids are known to reduce oxidative stress and reactive oxygen species (ROS) in cases of diabetes and hyperglycemia. The use of 70% ethanol as a solvent its effectiveness in extracting polar compounds and yielding diverse secondary metabolites. This study aimed to analyze the secondary metabolite content of the 70% ethanol extract of winged bean fruit and analyze antihyperglycemic activity in male mice induced
with alloxan. Tests were conducted on five groups: negative control Na CMC 0,5%; positive control glibenclamide 0.0208 mg/32g BW; and extract doses of 100, 300, and 500 mg/kg BW. Blood glucose levels were measured using the GOD-PAP method using photometer. Phytochemical screening revealed presence of alkaloids, flavonoids, tannins, and phenolics. The antihyperglycemic test showed extract
lowered blood glucose levels by 60,39%; 64,88%; and 69,71% at the respective doses. SPSS analysis showed normal distribution (p=0,457). One Way ANOVA indicated a significant difference (p=0,001), and Post Hoc Tamhane showed no significant difference between the positive control and all extract doses. The 100 mg/kg BW dose was as effective as glibenclamide.
