Abstract

Hiperglikemia adalah kondisi meningkatnya kadar glukosa darah yang dapat memicu resistensi insulin dan menjadi faktor risiko utama diabetes melitus (DM). Di Indonesia, prevalensi DM meningkat dari 6,9% pada 2013 menjadi 8,5% pada 2018. Pengobatan konvensional memiliki efek samping dan biaya tinggi, sehingga diperlukan alternatif pengobatan dari bahan alam. Salah satu tanaman yang berpotensi sebagai antihiperglikemik adalah daun rumput knop (Hyptis capitata Jacq.), yang diketahui mengandung senyawa aktif seperti alkaloid, flavonoid, steroid, dan tanin. Penelitian terhadap ekstrak etanol 96% daun rumput knop dilakukan pada mencit jantan yang diinduksi aloksan dosis 150 mg/kgBB dan pembebanan glukosa 5%. Lima kelompok mencit diberi perlakuan berbeda, yaitu kontrol negatif, kontrol positif, serta ekstrak daun rumput knop dengan dosis 250, 500, dan 700 mg/kgBB. Pengukuran kadar glukosa darah dilakukan secara berkala menggunakan fotometer, dan data dianalisis menggunakan program statistik SPSS. Hasil penelitian menunjukkan bahwa pemberian ekstrak etanol 96% daun rumput knop (Hyptis capitata Jacq.) memiliki aktivitas antihiperglikemik pada mencit putih jantan dengan dosis efektif adalah dosis ekstrak 250 mg/kg BB yang tidak berbeda bermakna dengan kontrol positif berdasarkan uji One Way ANOVA dan uji post-hoc Tamhane’s./ Hyperglycemia is a condition characterized by elevated blood glucose levels, which can trigger insulin resistance and serve as a major risk factor for diabetes mellitus (DM). In Indonesia, the prevalence of DM increased from 6.9% in 2013 to 8.5% in 2018. Conventional treatments often have side effects and are costly, thus alternative treatments derived from natural sources are needed. One plant with potential antihyperglycemic properties is knob grass leaves (Hyptis capitata Jacq.), which are known to contain active compounds such as alkaloids, flavonoids, steroids, and tannins. A study using 96% ethanol extract of knob grass leaves was conducted on male mice induced with alloxan at a dose of 150 mg/kgBW and loaded with 5% glucose. The mice were divided into five groups receiving different treatments: negative control, positive control, and knob grass leaf extract at doses of 250, 500, and 700 mg/kgBW. Blood glucose levels were measured periodically using a photometer, and the data were analyzed using SPSS statistical software. The results showed that administration of the 96% ethanol extract of knob grass leaves (Hyptis capitata Jacq.) exhibited antihyperglycemic activity in male white mice, with the effective dose being 250 mg/kgBW, which showed no significant difference compared to the positive control based on One-Way ANOVA and Tamhane’s post-hoc test
