Kolesterol merupakan suatu senyawa lemak kompleks. Hiperkolesterol merupakan kondisi meningkatnya kadar kolesterol melebihi batas normal yaitu >200 mg/dl. Obesitas merupakan kondisi terjadinya penimbunan lemak ditubuh yang berpengaruh pada kadar profil lipid. Tujuan penelitian ini untuk mengetahui aktivitas antikolesterol dan antiobesitas pada mencit jantan, mengetahui dosis yang efektif serta mengetahui kandungan senyawa total flavonoid dan fenolik dari ekstrak daun gedi. Daun gedi di ekstraksi dengan metode maserasi. Pengujian antikolesterol dan antiobesitas dilakukan menggunakan 36 ekor mencit yang dibagi dalam 6 kelompok yaitu kontrol negatif, kontrol positif simvastatin, kontrol positif orlistat, ekstrak 100 mg/kgBB, 300 mg/kgBB ekstrak 500 mg/kgBB yang diinduksi dengan prophylthioracil dan pakan tinggi lemak selama 16 hari. Hasil pengujian diperoleh % penurunan kadar kolesterol pada dosis 500 mg/kgBB (31%), simvastatin (26%), dosis 300 mg/kgBB (25%) dan dosis 100 mg/kgBB (24%). Hasil pengujian diperoleh % penurunan berat badan pada dosis 100 mg/kgBB (4,90%), dosis 500 mg/kgBB (4,53%), dosis 300 mg/kgBB (4,44%) dan orlistat (4,38%). Ekstrak efektif sebagai antikolesterol namun kurang efektif sebagai antiobesitas dengan hasil analisis data ANOVA menunjukkan nilai p (sig) < 0,05 dan MANOVA p (sig) < 0,05. Dosis efektif ditunjukkan pada dosis 100 mg/kgBB. Hasil flavonoid total 91,646 mg/QE/g ekstrak dan hasil fenolik total 1,025 mg/GAE/g ekstrak. / Cholesterol is a complex fatty compound. The purpose of this study was to determine the anticholesterol and antiobesity activity in male mice, determine the effective dosage and determine the total content of flavonoid and phenolic compounds from gedi leaf extract. Gedi leaves are extracted by the maceration method. Anticholesterol and antiobesity testing was carried out using 36 mice divided into 6 groups, namely negative control, positive control of simvastatin, positive control of orlistat, extract of 100 mg/kgBB, 300 mg/kgBB of extract of 500 mg/kgBB induced with prophylthioracil and high-fat feed for 16 days. The test results were obtained a % reduction in cholesterol levels at a dose of 500 mg/kgBB (31%), simvastatin (26%), a dose of 300 mg/kgBB (25%) and a dose of 100 mg/kgBB (24%). The test results were obtained % weight loss at a dose of 100 mg/kgBB (4.90%), a dose of 500 mg/kgBB (4.53%), a dose of 300 mg/kgBB (4.44%) and orlistat (4.38%). The extract was effective as an anticholesterol but less effective as an antiobesity with the results of ANOVA data analysis showing a p (sig) value of < 0.05 and MANOVA p (sig) < 0.05. The effective dose is indicated at a dose of 100 mg/kgBB. The total flavonoid yield was 91.646 mg/QE/g extract and the total phenolic yield was 1.025 mg/GAE/g extract.
