Abstract

Acne vulgaris adalah penyakit kulit inflamasi kronis yang umum terjadi, terutama pada remaja dan dewasa, dengan prevalensi di Indonesia mencapai 85–100%. Penyebab utamanya adalah infeksi bakteri Propionibacterium acnes yang memicu peradangan pada unit pilosebasea. Penggunaan antibiotik sering menjadi terapi utama, namun pemakaian tidak rasional dapat menimbulkan resistensi. Oleh karena itu, dibutuhkan alternatif pengobatan yang aman seperti bahan alami. Daun biwa (Eriobotrya japonica) mengandung senyawa metabolit sekunder seperti alkaloid, flavonoid, tanin, kuinon, saponin, dan steroid yang berpotensi sebagai antibakteri. Penelitian ini bertujuan mengetahui aktivitas antibakteri ekstrak etanol 96% daun biwa serta menentukan nilai Konsentrasi Hambat Minimum (KHM) dan Konsentrasi Bunuh Minimum (KBM) terhadap P.acnes. Metode yang digunakan yaitu uji sumuran dengan vankomisin sebagai kontrol positif dan DMSO 10% sebagai kontrol negatif. Zona hambat terbaik diperoleh pada konsentrasi 50.000 ppm sebesar 16 mm. Uji KHM menggunakan spektrofotometri UV-Vis tidak menunjukkan inhibisi >80%, sedangkan uji KBM melalui metode spread plate menunjukkan tidak adanya pertumbuhan bakteri pada kontrol negatif. Hal ini disebabkan oleh kontaminasi atau kesalahan teknis saat inokulasi pada media MHA./ Acne vulgaris is a common chronic inflammatory skin disease, especially in adolescents and adults, with a prevalence in Indonesia reaching 85–100%. The main cause is Propionibacterium acnes bacterial infection that triggers inflammation of the pilosebaceous unit. The use of antibiotics is often the main therapy, but irrational use can lead to resistance. Therefore, safe alternative treatments are needed such as natural ingredients. Loquat leaves (Eriobotrya japonica) contain secondary metabolites such as alkaloids, flavonoids, tannins, quinones, saponins, and steroids that have antibacterial potential. This study aims to determine the antibacterial activity of 96% ethanol extract of loquat leaves and determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) values against P. acnes. The method used was a well test with vancomycin as a positive control and 10% DMSO as a negative control. The best inhibition zone was obtained at a concentration of 50,000 ppm of 16 mm. The MIC test using UV-Vis spectrophotometry showed no inhibition >80%, while the MBC test using the spread plate method showed no bacterial growth in the negative control. This was caused by contamination or technical errors during inoculation into MHA media
