Abstract
Jerawat terjadi akibat penumpukan minyak memicu peradangan dan aktivitas bakteri Propionibacterium acnes. Penggunaan bahan alam seperti daun biwa mengandung senyawa metabolit sekunder seperti flavonoid, tanin, fenolik menjadi alternatif terapi antibakteri yang aman. Untuk meningkatkan efektivitas antibakteri, ekstrak biwa dimodifikasi perak nitrat (AgNO3) konsentrasi 1.000 ppm dalam empat variasi rasio ekstrak : AgNO3 (0,25:1; 0,5:1; 1:1; 2:1) dengan pengadukan kontinu tiap 6 jam selama 4 hari, dan dikonfirmasi absorbansi dengan Spektrofotometer UV-Vis 200-600 nm. Modifikasi perak nitrat melepaskan ion Ag⁺ yang efektif merusak dan menembus membran bakteri, sementara senyawa aktif ekstrak biwa dapat meningkatkan efek antibakteri. Aktivitas antibakteri ekstrak daun biwa termodifikasi perak nitrat diuji menggunakan metode well diffusion. Antibiotik vankomisin digunakan sebagai kontrol positif dan DMSO 10% sebagai kontrol negatif. Aktivitas antibakteri ekstrak daun biwa termodifikasi perak nitrat 1.000 ppm (V1-V4) menunjukkan zona hambat 9,65-10,65 mm dengan kategori resistant. Meski demikian, nilai tersebut lebih tinggi dibandingkan ekstrak biwa murni yang tidak menunjukkan zona hambat (0 mm). Uji KHM dengan makrodilusi dan konfirmasi menggunakan spektrofotometer UV-Vis, meskipun hasilnya tidak dapat dianalisis lebih lanjut karena nilai inhibisi tidak valid. Uji KBM ekstrak daun biwa termodifikasi perak nitrat pada konsentrasi dibawah 1.000 ppm (500-31,25 ppm) tidak menunjukkan kemampuan membunuh bakteri Propionibacterium acnes. / Acne is an inflammatory skin condition caused by excess oil accumulation and the activity of Propionibacterium acnes. Biwa leaves contain secondary metabolites such as flavonoids, tannins, and phenolics, which are known to have antibacterial potential. To enhance antibacterial activity, the biwa leaf extract was modified using silver nitrate (AgNO₃) at 1,000 ppm in four different extract-to-AgNO₃ ratios (0.25:1; 0.5:1; 1:1; 2:1), stirred every 6 hours for 4 days. The modification was confirmed by UV-Vis spectrophotometry at 200–600 nm. Silver ions (Ag⁺) are known to disrupt bacterial membranes, and the presence of active compounds from the extract may enhance the antibacterial effect. The antibacterial activity was tested using the well diffusion method. Vancomycin and 10% DMSO were used as positive and negative controls, respectively. The modified extracts at 1,000 ppm (V1–V4) produced inhibition zones of 9.65–10.65 mm, classified as resistant but higher than the unmodified extract, which showed no inhibition. The MIC test was performed but could not be analyzed further due to invalid inhibition values. Meanwhile, the MBC test showed that concentrations below 1,000 ppm (500–31.25 ppm) failed to eliminate Propionibacterium acnes, indicating that the extract exhibited limited bacteriostatic activity.
