Abstract
Daun afrika (Gymnanthemum amygdalinum (Del) Sch.Bip.) merupakan tanaman yang banyak digunakan masyarakat Indonesia dengan tujuan pengobatan. Ekstrak etanol 96% daun afrika afrika (G. amygdalinum) terbukti memiliki aktivitas sebagai antihiperurisemia pada dosis 100, 200, dan 400 mg/kgBB. Penelitian ini bertujuan untuk mengetahui efek toksik yang dilakukan dengan penentuan nilai LD50, serta dilanjutkan dengan uji histopatologi organ. Ekstrak daun afrika (G. amygdalinum) dibuat dengan metode maserasi menggunakan pelarut etanol 96% dan hewan uji mencit jantan (Mus musculus). Pengujian toksisitas akut menggunakan dosis 300mg/kgBB dan 2000mg/kgBB, serta CMC-Na sebagai kontrol negatif. Dalam penentuan nilai LD50 dilakukan sesuai metode Organitation for Economic Co-operation Development 423 (OECD 423) mengacu pada Globally Harmonized Classification System (GHCS) dalam Pedoman uji praklinik BPOM 2022. Data yang didapatkan berupa kuantitatif dan kualitatif. Hasil yang didapatkan dalam penelitian, kelompok kontrol negatif, dosis 300mg/kgBB, dan 2000mg/kgBB tidak menunjukkan gejala toksisitas ataupun ditemukan kematian, sehingga LD50 ekstrak etanol 96% daun afrika masuk dalam golongan V pada GHCS yang berarti tidak terklasifikan dengan dosis toksik >5000 mg/kgBB. Pada pengamatan histopatologi terjadi sedikit infiltrasi limfosit pada organ hati di semua dosis perlakuan, serta degenerasi hepatosit pada organ hati kelompok 300mg/kgBB batch 3 dan kelompok 2000mg/kgBB, pada organ ginjal dan jantung tidak ditemukan kelainan. / African leaf (Gymnanthemum amygdalinum (Del) Sch.Bip.) is a plant that is widely used by Indonesian people for medicinal purposes. The 96% ethanol extract of African leaves (G. amygdalinum) is proven to have activity as antihyperuricemia at doses of 100, 200, and 400 mg/kgBB. This study aims to determine the toxic effects carried out by determining the LD50 value, and continued with organ histopathology tests. African leaf extract (G. amygdalinum) was prepared by maceration method using 96% ethanol solvent and male mice (Mus musculus) were tested. Acute toxicity testing uses doses of 300mg/kgBB and 2000mg/kgBB, and CMC-Na as a negative control. In determining the LD50 value, it is carried out according to the Organization for Economic Co-operation Development 423 (OECD 423) method referring to the Globally Harmonized Classification System (GHCS) in the BPOM preclinical test guidelines 2022. The data obtained are quantitative and qualitative. The results obtained in the study, the negative control group, the dose of 300mg / kgBB, and 2000mg / kgBB did not show symptoms of toxicity or death, so that the LD50 of 96% ethanol extract of African leaves is included in group V on GHCS which means it is not classified with toxic doses> 5000 mg / kgBB. Histopathological observations showed a slight infiltration of lymphocytes in the liver in all treatment doses, as well as hepatocyte degeneration in the liver of the 300mg/kgBB batch 3 group and the 2000mg/kgBB group, while no abnormalities were found in the kidneys and heart.
