Abstract
Lumbricus rubellus merupakan cacing tanah yang dikenal memiliki banyak manfaat baik dalam bidang kesehatan maupun agrikultur. Pengaruh menguntungkan yang diberikan L. rubellus diduga dipengaruhi oleh mikrobiota yang berada dalam usus cacing tanah tersebut. Namun demikian, studi mengenai komunitas mikroba terutama bakteri dalam usus L. rubellus masih sangat terbatas. Salah satu studi yang mempelajari tentang material genetik yang diambil langsung dari lingkungan sekitar adalah metagenomik. Metode ini dapat digunakan untuk menganalisis seluruh komunitas bakteri pada usus L. rubellus tanpa proses kultur terlebih dahulu. Pada penelitian ini dilakukan ekstraksi DNA komunitas bakteri dari usus L. rubellus untuk kemudian diproses dengan menggunakan Illumina untuk proses sequencing dan analisis metagenomik. Berdasarkan hasil filter dari data mentah, didapatkan sebanyak 238 OTU bakteri pada usus L. rubellus. Jumlah OTU tersebut telah menggambarkan keseluruhan komunitas mikroba dalam usus Lumbricus rubellus. Dari berbagai macam komunitas bakteri yang ada, terdapat sepuluh bakteri yang mendominasi yaitu, filum Proteobacteria, Fusobacterium, Actinobacteria, Spirochaetes, Bacteriodetes, Firmicutes, Chlorobi, Deinococcus-Thermus dan Elusimicrobia. Dari kesepuluh filum tersebut, filum Proteobacteria dan Bacteriodetes memiliki peran penting dalam menjaga kesehatan dan kelangsungan hidup Lumbricus rubellus. Informasi mengenai komposisi dan kelimpahan bakteri pada Lumbricus rubellus ini diharapkan dapat memberi gambaran mengenai kemungkinan kontribusi mikrobiota cacing tanah terhadap kesehatan inang maupun manfaat yang diberikan cacing tanah dalam bidang kesehatan dan agrikultur./Lumbricus rubellus is an earthworm that is known to have many benefits both in the fields of health and agriculture. The beneficial effect of L. rubellus is thought to be influenced by microbiota in the intestines of the earthworm. However, studies of microbial communities, especially bacteria in the intestine of L. rubellus, are still very limited. One study that studies genetic material taken directly from the environment is metagenomic. This method can be used to analyze the entire bacterial community in the L. rubellus intestine without a culture process first. In this study DNA extraction of bacterial communities from L. rubellus intestine was then processed using Illumina for the sequencing and metagenomic analysis. Based on the filter results from the raw data, there were 238 OTU bacteria found in the intestine of L. rubellus. The number of OTU has described the whole microbial community in the intestines of L. rubellus. Of the various bacterial communities that exist, there are ten bacteria that dominate namely, phylum Proteobacteria, Fusobacterium, Actinobacteria, Spirochaetes, Bacteriodetes, Firmicutes, Chlorobi, Deinococcus-Thermus and Elusimicrobia. Of the ten phyla, the phylum Proteobacteria and Bacteriodetes have an important role in maintaining the health and survival of L. rubellus. Information on the composition and abundance of bacteria in L. rubellus is expected to be able to give an idea of the possible contribution of earthworm microbiota to host health and the benefits that earthworms provide in health and agriculture.
