Abstract
Chitin or poly (β-(1-4)-N-acetyl-D-glucosamine) can be found abundantly in shrimp shells. Chitin can further be processed to produce N-acetylglucosamine (NAG), one of the forms of glucosamine that has many uses in the medicine and nutrition fields. NAG production can be done by fermentation method with the help of chitinolytic microorganisms. One of the chitinolytic microorganisms that has high chitinolytic activity is Providencia stuartii. Cell immobilization technique such as entrapment offers higher stability and efficiency. However, the study of Providencia stuartii cells immobilization using Structural Fibrous Network (SFN) of papaya trunk wood to produce NAG by fermentation has not been done. This research was conducted to determine the optimum SFN of papaya trunk wood size, SFN of papaya trunk wood : growth medium ratio and fermentation cycle for the production of NAG from the shrimp shells powder using submerged fermentation method. The research used experimental method with different sizes of matrix (1 x 1, 1.5 x 1.5 and 2 x 2 cm), different matrix : growth medium ratio (1:10, 1:15 and 1:20) and 4 fermentation cycles. SFN of papaya trunk wood with the size of 1 x 1 cm and SFN of papaya trunk wood : growth medium ratio of 1:10 were found to produce the optimum NAG concentration (238177.78±3153.48 ppm) also it yielded the highest NAG concentration for 238.18±3.15%, therefore were further used for the second stage research. The highest NAG production was in the first fermentation cycle (244266.67±20929.25 ppm) and NAG concentrations decreased over the last three cycles. Overall, SFN of papaya trunk wood can be used multiple times in NAG production. / Kitin atau poli (β-(1-4)-N-asetil-D-glukosamin) banyak ditemukan di dalam kulit udang. Kitin dapat digunakan untuk memproduksi N-asetilglukosamin (NAG) yang merupakan salah satu bentuk dari glukosamin yang memiliki banyak kegunaan dalam bidang kesehatan dan nutrisi. Produksi NAG dapat dilakukan dengan metode fermentasi menggunakan bantuan mikroorganisme kitinolitik. Salah satu mikroorganisme kitinolitik yang memiliki aktivitas kitinolitik yang tinggi adalah Providencia stuartii. Teknik imobilisasi sel secara entrapment meningkatkan stabilitas dan efektivitas. Namun, penelitian tentang imobilisasi sel Providencia stuartii menggunakan batang pohon papaya untuk menghasilkan NAG dengan metode fermentasi belum dilakukan. Oleh karena itu, penelitian ini dilakukan untuk menentukan ukuran optimal dari batang papaya, perbandingan batang papaya : media pertumbuhan, dan siklus fermentasi untuk memproduksi NAG dari fermentasi tepung kulit udang dengan metode fermentasi. Penelitian ini menggunakan metode eksperimental dengan menggunakan ukuran batang papaya yang berbeda (1 x 1, 1,5 x 1,5, dan 2 x 2 cm), perbandingan batang papaya : media pertumbuhan yang berbeda (1:10, 1:15, dan 1:20), dan 4 siklus fermentasi. Ukuran batang pepaya dan perbandingan batang papaya : media pertumbuhan yang menghasilkan hasil NAG optimum (238.177,78±3.153,48 ppm) adalah ukuran 1 x 1 cm dan perbandingan 1:10 dan juga menghasilkan rendemen NAG tertinggi sebesar 238.18±3.15%, sehingga dipakai untuk penelitian tahap kedua. Produksi NAG tertinggi terdapat pada siklus fermentasi pertama (244.266,67±20.929,25 ppm) dengan penurunan produksi NAG pada ketiga siklus berikutnya. Secara keseluruhan, batang papaya dapat digunakan untuk produksi NAG berulang kali.
