Abstract

Bacillus subtilis dan Bacillus megaterium merupakan bakteri resisten logam berat yang mampu bertindak sebagai agen bioreduksi cemaran logam berat dalam biota laut, seperti udang jerbung, cumi-cumi, dan kerang hijau yang diperoleh dari Tempat Pelelangan Ikan (TPI) Cituis, Tempat Pelelangan Ikan (TPI) Muara Angke, dan wilayah perairan Singaraja (Indramayu). Penelitian sebelumnya menunjukkan bahwa Bacillus subtilis dan Bacillus megaterium memiliki kemampuan dalam menurunkan kadar akumulasi cemaran logam berat. Untuk memperoleh hasil penurunan logam berat secara lebih maksimal, digunakan metode konsorsium bakteri resisten logam berat Cd, Cr, Cu, dan Pb dengan menggunakan 9 variasi level rasio perbandingan konsorsium. Sebelum ditentukan rasio perbandingan konsorsium terbaik yang merupakan tujuan dari penelitian ini, setiap jenis sampel yang diperoleh dari tiga lokasi berbeda dilakukan pengujian kandungan logam berat awal untuk menentukan nilai konsentrasi logam berat tertinggi pada sampel udang jerbung, cumi-cumi, dan kerang hijau. Variasi lokasi pengambilan sampel memberikan pengaruh yang signifikan terhadap jumlah logam berat Cu dan Pb. Kandungan logam berat Cu dan Pb tertinggi untuk setiap jenis sampel memiliki nilai persentase penurunan tertinggi dengan penggunaan rasio perbandingan konsorsium Bacillus subtilis dan Bacillus megaterium sebesar 20%:80%. Hasil tersebut selaras dengan ketiadaan zona bening di sekeliling sumur uji yang menandakan tingkat resistensi yang baik terhadap polutan logam berat yang sesuai dengan jumlah total bakteri resisten logam berat yang tumbuh. / Bacillus subtilis and Bacillus megaterium are the example of heavy metal resistant bacteria that are able to act as bioreduction agents for heavy metals contamination in marine samples, such as white shrimp, squid, and green mussels that are obtained from Cituis Fish Auction Place, Muara Angke Fish Auction Place, and Singaraja (Indramayu) area. Previous studies have shown that both Bacillus subtilis and Bacillus megaterium were capable to reduce the accumulation of heavy metals contamination. In order to obtain the maximum results of heavy metals Cd, Cr, Cu, and Pb reduction, consortium of heavy metal resistant bacteria method is being used in this research by using 9 variation levels of the consortium ratio. Before determining the best level of consortium ratio, as the aim of this research, each type of sample that were obtained from three different locations was tested for the amount of initial heavy metals content to determine the highest heavy metal concentration values in white shrimp sample, squid sample, and green mussels sample. Variation in sampling locations show a significant impact of the initial amount of heavy metals Cu and Pb. The highest content of heavy metals Cu and Pb for each type of sample has the highest percentage of heavy metals reduction by using 20%:80% ratio of Bacillus subtilis and Bacillus megaterium. These results are consistent with the absence of clear zones around the test wells which show the high resistance to heavy metals and compatibility with the amount of total heavy metal resistant bacteria that were grown.
