Abstract

Penyakit kardiovaskuler menjadi penyebab utama kematian secara global, salah satunya adalah terjadinya penggumpalan darah yang menyumbat aliran darah. Terapi dengan obat antikoagulan oral yang menjadi pilihan utama masih memiliki kekurangan terkait efek samping dan harga obat yang tergolong mahal. Oleh karena itu, perlu dikembangkan pengobatan alternatif yang tergolong murah dan aman, yaitu enzim yang bersifat fibrinolitik dari bakteri pada rusip, produk fermentasi ikan teri (Stolephorus sp) asal Bangka Belitung. Dalam makanan fermentasi yang terbuat dari Stolephorus sp sering ditemukan Bacillus subtilis. Enzim ekstraselulernya diketahui mempunyai potensi fibrinolitik. Oleh karena itu, diperlukan studi lebih lanjut sifat fibrinolitik tersebut. Perlu dicari bakteri yang memiliki sifat proteolitik dan selanjutnya diuji fibrinolitik dari bakteri tersebut yakni uji degradasi gumpalan darah, degradasi gumpalan euglobulin, dan degradasi fibrin. Selanjutnya, identifikasi bakteri menurut flowcharts Bergey’s Manual untuk mengetahui jenis bakteri yang didapatkan. Hasil pengujian menunjukkan bahwa kedelapan protein sampel dari bakteri rusip mempunyai potensi yang bersifat fibrinolitik dalam mendegradasi gumpalan darah, beberapa pita komponen penyusun fraksi euglobulin, dan degradasi fibrin secara langsung. Degradasi pita fraksi euglobulin dan fibrin divisualisasi melalui SDS-PAGE. Berdasarkan pada hasil identifikasi bakteri, kedelapan bakteri diprediksi berasal dari spesies Bacillus subtilis. / Cardiovascular disease is the main cause of death globally, one of which is blood clots blocking blood flow. Oral anticoagulant drugs as the main choice for therapy still have drawbacks related to the side effects and the price of drugs is quite expensive. Therefore, an alternative relatively cheap and safe treatment needs to be developed, namely fibrinolytic enzyme from bacteria in rusip, a fermented product of anchovy (Stolephorus sp) from Bangka Belitung. In fermented foods made from Stolephorus sp, Bacillus subtilis is often found. Its extracellular enzymes are known to have great potential for fibrinolytic activity. Therefore, further studies of fibrinolytic properties are needed. It is necessary to look for bacteria that have proteolytic properties and then test for fibrinolytic from these bacteria, namely blood degradation tests, euglobulin degradation, and fibrin degradation. Next, identify bacteria according to Bergey's Manual flowcharts to determine the type of bacteria obtained. The test results showed that the eight sample proteins from rusip bacteria had fibrinolytic potential in degrading blood clots, several bands of components of the euglobulin fraction, and direct fibrin degradation. The band degradation of the euglobulin and fibrin fractions was visualized using SDS-PAGE. According to the result of bacteria identification, the eight bacteria are predicted belong to the Bacillus subtilis species.
