Abstract
Susu merupakan salah satu produk konsumsi sehari-hari di seluruh dunia, yang banyak mengandung bakteri asam laktat. Bakteri asam laktat dapat berperan dalam membantu menjaga kesehatan manusia, sebagai kultur starter atau probiotik dalam industri susu. Namun, eksplorasi bakteri asam laktat dari susu sapi lokal belum banyak dilakukan di Indonesia. Penelitian ini melanjutkan penelitian sebelumnya dimana, 2 dari 36 isolat dari susu sapi lokal telah teridentifikasi. Namun, 34 isolat yang tersisa masih belum diketahui. Penelitian ini mengidentifikasi 2 dari 34 isolat yang belum diketahui yaitu, isolat ASY95 dan ASY98. Metode identifikasi yang digunakan meliputi identifikasi morfologi koloni dan sel, uji katalase, uji fermentasi gula, dan identifikasi molekuler DNA gen 16S rRNA. Isolat ASY95 dan ASY98 memiliki bentuk punctiform, elevasi convex, margin entire, berwarna putih, merupakan bakteri Gram positif dan katalase negatif, serta dapat memfermentasi gula fruktosa, laktosa, maltosa, sukrosa, dan dekstrosa. Berdasarkan identifikasi molekuler DNA gen 16S rRNA isolat ASY95 dan ASY98 teridentifikasi sebagai Lactobacillus paracasei subsp. paracasei. / Lactic acid bacteria can commonly be found in milk as one of daily main consumption products around the world. In dairy industry, lactic acid bacteria are used as starter culture or probiotics, which is known for its contribution in improving human health. However, little study has been done to explore lactic acid bacteria from local cow milk in Indonesia. This study was, thus, carried out as a continuation of previous study, where 2 out of 36 isolates from local cow milk has been identified. As 34 remaining isolates are still unknown, this study aimed to identify 2 out of 34 unknown isolates, namely isolate ASY95 and ASY98. Several methods were used in this study, including colony and cell morphology identification, catalase test, fermentation assay, and DNA gene 16S rRNA molecular identification. It was observed that both isolates have punctiform shape, convex elevation, entire margin, and white color, and also Gram positive and catalase negative bacteria. Based on the DNA gene 16S rRNA molelcular identification, isolate ASY95 and ASY98 were identified as Lactobacillus paracasei subsp. paracasei.
