Abstract
Penelitian yang dilakukan oleh peneliti sebelumnya menyebutkan bahwa daun
cempedak memiliki nilai hambat atau MIC pada bakteri E. coli dengan dosis
150mg/mL, maka dengan ini dilakukan untuk mengetahui aktivitas antibakteri dari
ekstrak daun cempedak terhadap bakteri E. coli dan S. pyogenes. Metode yang
dilakukan adalah mengukur zona hambat yang terbentuk dari metode sumuran atau
well diffusion. Daun cempedak dimaserasi dengan etanol absolut kemudian
dilakukan proses rotavapor dan pengeringan diatas penangas air untuk
mendapatkan ekstrak kering. Ekstrak kering dibuat beberapa konsentrasi yaitu 0,1
g/mL, 0,2 g/mL, 0,4 g/mL, 0,8 g/mL, 1,6 g/mL, 3,2 g/mL dengan kontrol positif
injeksi gentamisin sulfat dengan konsentrasi 10rb dan kontrol negatif DMSO. Pada
E. coli diujikan dua kali pengulangan (0,4 g/mL, 0,8 g/mL, 1,6 g/mL, 3,2 g/mL,
kontrol positif dan negatif). Untuk S. pyogenes diujikan tiga kali pengulangan untuk
konsentrasi 0,4 g/mL, 0,8 g/mL, 1,6 g/mL, 3,2 g/mL, dan dua kali untuk konsentrasi
0,1 g/mL, dan 0,2 g/mL. Pada skrining crude ekstrak etanol terdapat senyawa
flavonoid, fenol, dan tannin yang dapat menghambat aktivitas bakteri, sehingga
terdapat zona hambat pada bakteri S. pyogenes. Hasil penelitian menunjukan bahwa
crude ekstrak etanol daun cempedak dapat menghambat bakteri S. pyogenes dengan
rata-rata diameter zona hambat paling besar 11,49 mm pada konsentrasi 3,2 g/mL
tetapi tidak dapat menghambat bakteri E. coli/Previous research stated that cempedak leaf has an inhibitory value or MIC on E.
coli bacteria at a dose of 150mg/mL. This study was conducted to determine the
antibacterial activity of cempedak leaves extract against E. coli and S. pyogenes
bacteria. The research was conducted to measure the inhibition zone formed by
extract with well diffusion method. Cempedak leaves were macerated with absolute
ethanol and then the rotavapor process was carried out and drying on a water bath
to obtain a dry extract. The dry extract was made in several concentrations of 0.1
g/mL, 0.2 g/mL, 0.4 g/mL, 0.8 g/mL, 1.6 g/mL, 3.2 g/mL with a positive control is
gentamicin sulphate injection with a concentration of 10 thousand ppm and a
negative control DMSO. E. coli was tested twice (0.4 g/mL, 0.8 g/mL, 1.6 g/mL, 3.2
g/mL, positive and negative control). S. pyogenes was tested three times for
concentrations of 0.4 g/mL, 0.8 g/mL, 1.6 g/mL, 3.2 g/mL, and twice for
concentrations of 0.1 g/mL. and 0.2 g/mL. In screening crude ethanol extract there
are flavonoid, phenol, and tannin compounds that can inhibit bacterial activity, so
that there is an inhibition zone for S. pyogenes bacteria. The results showed that
crude ethanol extract of cempedak leaves could inhibit S. pyogenes bacteria with
an average diameter of the largest inhibition zone is 11.49 mm at a concentration
of 3,200 million ppm but could not inhibit E. coli bacteria.
