Abstract

Banyaknya tumpukan sampah yang ada pada lingkungan sekitar dapat menimbulkan berbagai macam dampak negatif, seperti bencana banjir maupun timbulnya berbagai macam penyakit yang berbahaya. Salah satu cara yang cukup efektif dalam mengurangi sampah yang ada, yaitu dengan menggunakan metode pengomposan. Terdapat salah satu metode pengomposan yang cukup mudah, tidak menghabiskan banyak biaya, tidak menimbulkan bau dan dapat dilakukan oleh siapapun di rumah. Metode ini dikenal dengan metode Takakura dengan menggunakan larutan starter EM4 & MOL. Dalam proses pengomposan, terdapat berbagai macam faktor yang berperan penting dan salah satunya karena adanya aktivitas metabolisme mikroorganisme yang bekerja di dalamnya. Proses sampai terbentuknya kompos harus melalui berbagai macam fase, dan fase yang paling penting dalam menentukan keberhasilan cepat atau lambat sampai terbentuk kompos adalah fase termofilik. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi mikroorganisme termofilik yang bekerja pada kompos selama fase termofilik dan melihat enzim yang dihasilkan. Mikroorganisme diisolasi pada fase termofilik saat rentang suhu pada kompos mencapai 43℃-45℃, kemudian dilakukan dilusi berseri pada sampel dari 10¹ - 10⁶ dan sampel di-spread pada medium NA & GYP. Sampel diinkubasi selama 24 jam pada suhu 55℃. Hasil penelitian menunjukan, didapatkan 9 isolat bakteri termofilik dengan identitas 5 isolat bakteri berbentuk basil, Gram negatif ; 3 isolat bakteri berbentuk basil, Gram positif ; dan 1 isolat bakteri berbentuk kokus, Gram positif. Berdasarkan flowchart uji biokimia pada Bergey’s Manual Of Determinative Bacteriology Ninth Edition, 5 isolat bakteri berbentuk basil, Gram negatif merupakan kelompok bakteri dengan genus Aeromonas ; 3 isolat bakteri berbentuk basil, Gram positif merupakan kelompok bakteri dengan genus Bacillus ; dan 1 isolat bakteri berbentuk kokus, Gram positif merupakan kelompok bakteri dengan genus Staphylococcus. Ke-9 isolat bakteri termofilik yang berhasil diisolasi dari kompos berperan dalam mempercepat perombakan dari bahan organik melalui proses metabolisme dalam tubuhnya. / The amount of waste in the surrounding environment can cause various kinds of negative impacts, such as floods and the emergence of various kinds of dangerous diseases. One way that is quite effective in reducing existing waste is by using the composting method. There is one method of composting that is quite easy, low of cost, does not cause odor and can be done by anyone at home. This method is known as the Takakura method with EM4 & MOL solutions. In the composting process, there are various factors that play an important role and one of them is the activity of microorganisms that work in it. The process until the formation of compost must go through with various phases, and the most important phase in determining the success of fast or slowness of composting process is in the thermophilic phase. This research purpose is to isolate and identify the thermophilic microorganisms that work on compost during the thermophilic phase and see the various products of enzymes by thermophilic microorganisms. Microorganisms were isolated in the thermophilic phase when the compost temperature range reached 43℃-45℃, then serial dilution was carried out on the sample from 10¹ - 10⁶ and the sample was spread on NA & GYP medium. Samples were incubated for 24 hours at 55℃. The results showed that there were 9 isolates of thermophilic bacteria with the identity of 5 bacterial isolates in the form of bacilli, Gram negative; 3 bacterial isolates in the form of bacilli, Gram positive ; and 1 bacterial isolate in the form of cocci, Gram positive. Based on the flowchart of biochemical tests in Bergey's Manual Of Determinative Bacteriology Ninth Edition, 5 bacterial isolates were in the form of bacilli, Gram negative was a group of bacteria with the genus Aeromonas ; 3 bacterial isolates in the form of bacilli, Gram positive is a group of bacteria with the genus Bacillus ; and 1 bacterial isolate in the form of cocci, Gram positive is a group of bacteria with the genus Staphylococcus. The 9 isolates of thermophilic bacteria that were isolated from compost play a role in accelerating the breakdown of organic matter through metabolic processes in the body.
