Abstract
Pembuangan limbah tekstil yang tidak diregulasi dengan baik dapat menyebabkan pencemaran lingkungan dan membahayakan organisme yang hidup disekitarnya. Hal tersebut dapat dikarenakan sifat dari limbah pewarna yang beracun baik secara mutagenicity, carcinogenic, maupun sifat beracun lainnya. Oleh karena itu pencarian metode dekolorisasi yang terbaik perlu untuk dilakukan. Dekolorisasi secara biologi dapat menjadi salah satu solusi yang dapat digunakan untuk mengatasi permasalahan tersebut. Salah satu cara dalam dekolorisasi secara biologi adalah dengan menggunakan mikroorganisme hidup untuk dapat mendekolorisasi pewarna yang menjadi limbah. Salah satu mikroorganisme yang optimal untuk dapat melakukan dekolorisasi adalah yeast yang salah satunya dapat ditemukan dalam ragi tuak. Dekolorisasi dengan menggunakan ragi tuak dalam penelitian ini dilakukan dengan pertama mengisolasi ragi yang ada, kemudian dilakukan uji %dekolorisasi untuk mengetahui efektivitas isolat dalam mendekolorisasi, dan dilakukan pengujian biokimia dan morfologi untuk dapat menentukan genus dari isolat. Berdasarkan penelitian yang dilakukan maka didapati terdapat dua isolat yang berhasil untuk di isolasikan yaitu SS.CR.1 dan SS.CR.2. Ditemukan bahwa SS.CR.1 dapat mendekolorisasi pewarna MG dengan baik dan SS.CR.2 ditemukan dapat mendekolorisasi EB dengan baik. Selain dari itu juga ditemukan bahwa SS.CR.1 dan SS.CR.2 masuk kedalam genus dari antara Candida spp. ataupun Saccharomyces spp. tetapi dapat dipastikan kedua isolat bukan dari Candida albicans./ Disposal of textile dye waste that is not properly regulated can cause environmental pollution and endanger organisms that live around it. This can be due to the nature of the dye waste which is toxic in terms of mutagenicity, carcinogenicity, and other toxic properties. Therefore, it is necessary to search for the best decolorization method. Biological decolorization can be one solution that can be used to overcome these problems. One way of biological decolorization is to use live microorganisms to decolorize dyes that become waste. One of the optimal microorganisms to be able to decolorize is yeast, one of which can be found in tuak. Decolorization using tuak in this study was carried out by first isolating the yeast in tuak, then a %decolorization test was carried out to determine the effectiveness of the isolate in decolorizing, and biochemical and morphological tests were carried out to determine the genus of the isolate. Based on the research conducted, it was found that there were two isolates that were successfully isolated, namely SS.CR.1 and SS.CR.2. It was found that SS.CR.1 could decolorize MG dye well and SS.CR.2 was found to decolorize EB well. Apart from that, it was also found that SS.CR.1 and SS.CR.2 belonged to the genus Candida spp. or Saccharomyces spp. but it can be ascertained that the two isolates were not from Candida albicans.
