Abstract
Limbah pewarna dan logam berat menjadi salah satu penyebab pencemaran berbahaya bagi lingkungan. Diperlukan solusi sebagai upaya untuk mengatasi masalah pencemaran oleh kedua agen tersebut. Beberapa metode tersedia, tetapi tidak semua dapat menangani masalah dengan aman dan permanen. Potensi enzim dekolorisasi dari bakteri resisten tembaga dapat dimanfaatkan menjadi solusi alternatif masalah tersebut. Terdapat 3 sampel bakteri resisten tembaga yang tersedia namun belum diketahui peran enzim maupun potensi dekolorisasi pewarna terhadap lingkungan. Oleh karena itu, dilakukan penelitian untuk mengetahui karakterisasi dan identifikasi enzim yang berperan dalam proses dekolorisasi pewarna dari bakteri resisten tembaga tersebut. Dalam penelitian ini, pengujian potensi dekolorisasi dilakukan dengan metode streak di agar padat untuk mendapatkan data secara kualitatif dan pengukuran persentase dekolorisasi
untuk mendapatkan data secara kuantitatif. Profil protein ekstraseluler didapatkan melalui SDS-PAGE. Dilakukan juga GC-MS untuk mengetahui hasil pecahan metabolit pewarna yang dihasilkan. Hasil yang ada menunjukkan bahwa adanya
pita protein yang diperkirakan merupakan enzim dekolorisasi seperti azoreduktase dan peroksidase. Pita protein lain diperkirakan merupakan metalloproteinase, protein yang berperan dalam resistensi bakteri terhadap logam berat tembaga.

Dyes and copper heavy metal is one of the cause of hazardous pollution to enviroment. A solution is needed as an effort to resolve environmental problem caused by those 2 hazardous agent. Methods to solve them are available but, not all those methods can solve those problem in safe and permanent way. Potential of decolorization enzyme from copper resistant bacteria can be an alternative method to solve the problem. 3 sample of copper resistant bacteria is available but the role and potency of their enzyme is still unknown. Because of that, research is being conducted to learn more about characteristics and identification of role of enzyme in decolorization of dye drom those copper resistant bacteria. In this research, decolorization potency is tested by streak method on dye agar media to get qualitative data and measuring decolorization percentage to get quantitative data. Extracellular protein profiling is done by SDS-PAGE method. GC-MS is done to learn about the degradation result and metabolite. The result show that there is protein band that is predicted to be decolorization enzyme such as azoreductase and peroxidase. Other protein band is predicted to be metalloproteinase, protein that responsible for bacterial copper resistancy.
