Abstract

Tanaman pecut kuda (Stachytarpheta cayennensis (Rich.) Vahl) merupakan gulma atau tanaman liar yang tumbuh daerah berdataran iklim tropis dan subtropis yang belum banyak dimanfaatkan oleh masyarakat. Tanaman ini berpotensi sebagai antioksidan karena mengandung senyawa kimia seperti alkaloid, triterpenoid, flavanoid, saponin, dan tanin yang dapat digunakan sebagai antioksidan alami. Antioksidan merupakan salah satu senyawa yang dalam kadar rendah secara signifikan dapat mencegah terjadinya oksidasi substrat dalam reaksi rantai. Tujuan penelitian ini adalah untuk mengetahui formulasi optimal sediaan serum, aktivitas antioksidan, evaluasi mutu fisik dan stabilitas sediaan serum ekstrak etanol 96% daun pecut kuda (Stachytarpheta cayennensis (Rich.) Vahl)) yang memenuhi syarat. Ekstraksi daun pecut kuda (Stachytarpheta cayennensis (Rich.) vahl) dilakukan dengan menggunakan metode maserasi selama 3x24 jam dengan menggunakan pelarut etanol 96%. Aktivitas antioksidan diuji dengan menggunakan metode DPPH (1-1-difenil-2-pikrilhidrazil) yang dihitung menggunakan perhitungan nilai IC50 (inhibition concentration). Hasil estraksi daun pecut kuda (Stachytarpheta cayennensis) diperoleh kadar air ekstrak sebesar 13,16% dan rendemen ekstrak sebesar 11,8%, memiliki kandungan fitokimia yaitu alkaloid, flavanoid, tanin, saponin, dan triterpenoid, serta memiliki aktivitas antioksidan kuat dengan IC50 sebesar 46,57 µg/mL. Sediaan serum memenuhi persyaratan mutu fisik sediaan serta stabil dalam penyimpanan selama empat siklus adalah formula satu. Hasil evaluasi sediaan yang memenuhi syarat mutu adalah homogenitas, pH dengan rata-rata yaitu 5,71, bobot jenis dengan rata-rata yaitu 1,043 g/cm3, daya lekat dengan rata-rata yaitu 37,3 mikrodetik dan viskositas dengan nilai rata-rata yaitu 231,2 cp. Sedangkan pada mutu fisik sediaan daya sebar belum memenuhi syarat dengan rata-rata yaitu 9,83 cm. Hasil uji kestabilan dengan cycling test pada siklus 1-4 memenuhi persyaratan kecuali daya sebar./ Horse whip plant (Stachytarpheta cayennensis) is a weed or wild plant that grows in tropical and subtropical climates that have not been widely utilized by the community. This plant has potential as an antioxidant because it contains chemical compounds such as alkaloids, triterpenoids, flavanoids, saponins, and tannins that can be used as natural antioxidants. Antioxidant is one of the compounds that in low levels can significantly prevent the oxidation of substrates in chain reactions. The purpose of this study was to determine the optimal formulation of serum preparations, antioxidant activity, physical quality evaluation and stability of serum preparations of 96% ethanol extract of horse whip leaves (Stachytarpheta cayennensis) that meet the requirements. Extraction of horse whip leaves (Stachytarpheta cayennensis) was carried out using maceration method for 3x24 hours using 96% ethanol solvent. Antioxidant activity was tested using DPPH (1-1-diphenyl-2-picrylhydrazyl) method which was calculated using IC50 (inhibition concentration) value. The extraction results of horse whip leaves (Stachytarpheta cayennensis) obtained extract moisture content of 13.16% and extract yield of 11.8%, has phytochemical content of alkaloids, flavanoids, tannins, saponins, and triterpenoids, and has strong antioxidant activity with IC50 of 46.57 µg/mL. Serum preparation that meets the physical quality requirements of the preparation and is stable in storage for four cycles is formula one. The results of the evaluation of preparations that meet the quality requirements are homogeneity, pH with an average of 5.71, specific gravity with an average of 1.043 g/cm3, adhesion with an average of 37.3 microseconds and viscosity with an average value of 231.2 cp. While in the physical quality of the preparation, the spreadability has not met the requirements with an average of 9.83 cm. The results of the stability test with the cycling test in cycles 1-4 meet the requirements except for spreadability.
