Abstract

Tanaman sirsak (Annona muricata L.) dan daun kemangi (Ocimum x africanum L.) berpotensi sebagai inhibitor α-glukosidase. Tujuan umum dalam penelitian ini yaitu memanfaatkan daun sirsak dan daun kemangi dalam pembuatan minuman fungsional yang diharapkan memiliki aktivitas inhibisi α-glukosidase. Pada penelitian pendahuluan, daun sirsak diolah menjadi teh melalui proses pelayuan dan pengeringan, sedangkan daun kemangi diolah menjadi ekstrak dengan metode maserasi menggunakan pelarut etanol 96% . Daun teh sirsak dan ekstrak daun kemangi kemudian diolah menjadi minuman fungsional. Pembuatan minuman fungsional terdiri dari 3 level berat ekstrak 0,3; 0.5; 0,7 g dan 4 level konsentrasi pemanis yaitu 0, 1,5, 3, 4,5 g/L. Minuman fungsional dilakukan uji organoleptik (hedonik dan skoring) dan uji fisik (pH, warna dan total padatan terlarut) kemudian pada minuman fungsional terpilih dilakukan pengujian terhadap total fenolik, total flavonoid, aktivitas antioksidan, aktivitas inhibisi α-glukosidase, dan kinetika penghambatan enzim pada minuman fungsional. Total fenolik daun sirsak yaitu sebesar 60,18 ± 0,96 mg GAE/L, total flavonoid 5,12 ± 0,14 mg QE/g, aktivitas antioksidan (IC50) yaitu 46876,69 ± 670,68 ppm serta inhibisi α-glukosidase (IC50) yaitu 35806,25 ± 1774,34 ppm. Total fenolik kemangi yaitu 41,54 ± 0,48 mg GAE/g, total flavonoid 12, 11 ± 0,23 mg QE/g, aktivitas antioksidan (IC50) yaitu 189,22 ± 6,86 ppm serta aktivitas inhibisi α-glukosidase (IC50) yaitu 38,67± 1,91 ppm. Minuman terpilih yaitu dengan berat ekstrak 0,7 g dan konsentrasi pemanis 3g/L memiliki nilai hedonik keseluruhan rata – rata yaitu 5,19 dari skala 1-7, total kandungan fenolik yaitu 68,82 ±1,71 mg GAE/g, total flavonoid 30,97 ± 0,35 mg QE/g, aktivitas antioksidan (IC50) yaitu 13023,29 ± 248,63 ppm, aktivitas inhibisi α-glukosidase (IC50) yaitu 34289,77± 1573,99 ppm dan jenis kinetika inhibisi enzim minuman fungsional yaitu kompetitif.
