Abstract

Shrimps processing may result in huge amount of shrimp shells waste containing chitin that can further be processed into N-acetylglucosamine which has been extensively used to treat joint damage. N-acetylglucosamine production can be done through microbial fermentation by using the chitinolytic microorganisms. As a chitinolytic microorganism, Providencia stuartii may be utilized in the form of immobilized cells. Pumice is a porous and rigid stone that offers superior mechanical strength, making it suitable to be used for immobilization process. Hence, this research was conducted to determine the optimum pumice stone size, pumice stone:growth medium ratio (w/v), and fermentation cycle for the N-acetylglucosamine production through the fermentation of shrimp shells powder using Providencia stuartii cells immobilized pumice stones. The research used experimental method to conduct the submerged fermentation process with different pumice stone size, pumice stone:growth medium ratio (w/v), and fermentation cycle. The fermentation was carried out for four days at 37oC and pH of 7.0. Pumice stones of 1.0 x 1.0 x 1.0 cm with pumice stone:growth medium ratio (w/v) of 1:5 were found to be the optimum conditions which successfully immobilized 89.99±1.65% cells and produced 331366.67±7338.95 ppm N-acetylglucosamine (331.37±7.34% yield). Thus, these conditions were further used in the second stage research. The highest N-acetylglucosamine concentration of 322966.66±2465.30 ppm (322.97±2.46% yield). was obtained in the first fermentation cycle which then decreased and remained stable throughout the last three cycles. The stable yield of N-acetylglucosamine on the final three cycles indicated that pumice stone can be utilized as a stable and robust carrier for immobilization process. / Proses pengolahan udang akan menghasilkan limbah kulit udang dalam jumlah besar. Kulit udang tersebut mengandung kitin yang dapat diolah menjadi N-asetilglukosamin yang banyak digunakan untuk menjaga kesehatan persendian tulang. N-asetilglukosamin dapat diproduksi melalui fermentasi dengan menggunakan mikroorganisme kitinolitik. Sebagai salah satu mikroorganisme kitinolitik, Providencia stuartii dapat digunakan dalam bentuk terimobil. Batu apung merupakan batu berpori dan kuat sehingga membuatnya menjadi salah satu material yang cocok digunakan sebagai matriks imobilisasi, Oleh karena itu, penelitian ini dilakukan untuk menentukan ukuran batu apung optimum, rasio batu apung dan media pertumbuhan (b/v) optimum, serta siklus fermentasi optimum untuk produksi N-asetilglukosamin melalui fermentasi tepung kulit udang menggunakan batu apung yang terimobilisasi oleh Providencia stuartii. Penelitian menggunakan fermentasi media cair ini dilakukan secara eksperimental dengan ukuran batu apung, rasio batu apung dan media pertumbuhan, serta siklus fermentasi yang berbeda. Fermentasi dilakukan selama empat hari pada suhu 37oC and pH 7,0. Batu apung dengan ukuran 1,0 x 1,0 x 1,0 cm dan rasio batu apung:media pertumbuhan (b/v) 1:5 diperoleh sebagai kondisi optimum yang berhasil mengimobilisasi 89,99±1,65% sel serta menghasilkan 331366,67±7338,95 ppm N-asetilglukosamin (331.37±7.34% rendemen) sehingga digunakan pada penelitian tahap dua. Konsentrasi N-asetilglukosamin tertinggi sebanyak 322966,66±2465,30 ppm (322.97±2.46% rendemen) diperoleh pada siklus fermentasi pertama yang selanjutnya menurun dan akhirnya stabil pada tiga siklus terakhir. Hasil N-asetilglukosamin yang stabil pada siklus kedua hingga keempat menandakan bahwa batu apung dapat digunakan sebagai matriks imobilisasi.
