Abstract

Udang menghasilkan limbah berupa cangkang udang dalam jumlah yang cukup besar. Pada cangkang udang terkandung kitin yang dapat didegradasi menjadi senyawa N-asetilglukosamin melalui proses fermentasi dengan bantuan bakteri kitinolitik. Imobilisasi sel bakteri dapat dilakukan untuk meningkatkan produktivitas. Sel yang terimobilisasi diketahui memiliki stabilitas sel yang lebih baik dibandingkan dengan sel bebas. Salah satu bahan yang dapat digunakan untuk imobilisasi bakteri adalah spons loofah. Penelitian ini bertujuan untuk mengetahui kadar N-asetilglukosamin selama fermentasi berulang dengan sel bakteri Providencia stuartii terimobilisasi pada spons loofah, menentukan ukuran spons loofah terbaik, menentukan rasio antara spons loofah dengan media pertumbuhan terbaik, dan menentukan stabilitas imobilisasi sel Providencia stuartii pada fermentasi berulang. Ukuran spons loofah yang digunakan pada penelitian ini adalah 1,0 x 1,0 cm, 1,5 x 1,5 cm dan 2,0 x 2,0 cm dengan ketebalan berkisar antara 0,4-0,6 cm. Rasio antara spons loofah dengan media pertumbuhan yang digunakan adalah sebesar 1:200, 1:250 dan 1:300 (b/v). Fermentasi berlangsung selama 4 hari dengan substrat berupa tepung cangkang udang. Ukuran spons loofah terbaik yang diperoleh dari penelitian ini adalah ukuran 1,0 x 1,0 cm dan rasio terbaik adalah 1:200 (b/v). Rata-rata kadar N-asetilglukosamin yang dihasilkan selama fermentasi berulang adalah sebesar 215840,46±7663,95 ppm. Konsentrasi N-asetilglukosamin stabil selama fermentasi berulang hingga 4 siklus. / Shrimp produce high waste in the form of shrimp shells. Shrimp shells contain chitin that can be degraded to N-acetylglucosamine through fermentation process by chitinolytic bacteria. Cell immobilization can be done to increase productivity. Immobilized cells are known to have better cell stability compared to the free cell. One of the materials that can be used for bacterial cell immobilization is loofah sponge. This study aims to know concentration of N-acetylglucosamine during repeated fermentation cycle by Providencia stuartii cells immobilized on loofah sponge, determine the best size of loofah sponge, determine the best loofah sponge to growth medium ratio, and determine the stability of immobilization of Providencia stuartii cell during repeated fermentation cycle. Size of loofah sponge used in this study are 1,0 x 1,0 cm, 1,5 x 1,5 cm and 2,0 x 2,0 cm with thickness 0,4-0,6 cm. The loofah sponge to growth medium ratio used in this study are 1:200, 1:250 and 1:300 (w/v). Fermentation is conducted for 4 days with shrimp shell powder as the fermentation substrate. The best size of loofah sponge is 1,0 x 1,0 cm and the best ratio is 1:200 (b/v). Mean of N-acetylglucosamine concentration produced during repeated fermentation cycle is 215840,46±7663,95 ppm. Concentration of N-acetylglucosamine stable during repeated fermentation until 4 cycles.
